目的::明确IL-8与表皮生长因子( EGF)对黑素瘤细胞增殖的影响。方法:将黑素瘤细胞A375分为9组:EGF组;EGF+AG1478组;AG1478组;IL-8组;IL-8I组;IL-8+IL-8I组;EGF+ IL-8组;EGF+IL-8+AG1478+IL-8I组及对照组。采用MTT法检测各组细胞增殖情况;免疫印迹法检测EG-FR、蛋白激酶B、丝裂原活化蛋白激酶、磷酸化蛋白激酶B、磷酸化丝裂原活化蛋白激酶、磷酸化表皮生长因子受体的表达;实时荧光定量聚合酶链式反应检测EGFR与IL-8R mRNA水平的表达。结果:EGF组黑素瘤细胞增殖、上述蛋白的表达和mRNA水平显著高于对照组、EGF+AG1478组和AG1478组。IL-8组显著高于对照组、IL-8I组和IL-8+IL-8I组(均P<0.05),EGF+AG1478组显著高于EGF组和IL-8组( P<0.05)。结论: IL-8与EGF均能促进人黑素瘤细胞的增殖,它们之间可能具有相互协同作用,这种协同作用可能与AKT和MAPK等共同通路作用有关。

목적::명학IL-8여표피생장인자( EGF)대흑소류세포증식적영향。방법:장흑소류세포A375분위9조:EGF조;EGF+AG1478조;AG1478조;IL-8조;IL-8I조;IL-8+IL-8I조;EGF+ IL-8조;EGF+IL-8+AG1478+IL-8I조급대조조。채용MTT법검측각조세포증식정황;면역인적법검측EG-FR、단백격매B、사렬원활화단백격매、린산화단백격매B、린산화사렬원활화단백격매、린산화표피생장인자수체적표체;실시형광정량취합매련식반응검측EGFR여IL-8R mRNA수평적표체。결과:EGF조흑소류세포증식、상술단백적표체화mRNA수평현저고우대조조、EGF+AG1478조화AG1478조。IL-8조현저고우대조조、IL-8I조화IL-8+IL-8I조(균P<0.05),EGF+AG1478조현저고우EGF조화IL-8조( P<0.05)。결론: IL-8여EGF균능촉진인흑소류세포적증식,타문지간가능구유상호협동작용,저충협동작용가능여AKT화MAPK등공동통로작용유관。
Objective:To determine the effects of IL-8 and epidermal growth factor ( EGF) on the prolif-eration of melanoma cell. Methods: The human melanoma cells A375 were divided into 9 groups: control group, EGF group, EGF combined AG1478 group, AG1478 group, IL-8 group, IL-8I group, IL-8 com-bined IL-8I group, EGF combined IL-8 group and EGF combined IL-8, AG1478 and IL-8I group. The pro-liferation of melanoma cell was detected by MTT. The level of EGFR, protein kinase B ( Akt) , mitogen activa-ted protein kinase (MAPK), phosphorylated protein kinase B (P-Akt), phosphorylated mitogen activated protein kinase ( p-MAPK) and phosphorylated epidermal growth factor receptor ( p-EGFR) were detected by western blot. The expression of EGFR and IL-8R mRNA levels were detected by qT-PCR. Results:The cell proliferation, the levels of proteins above and EGFR, and IL-8R mRNA levels in EGF group were higher than that in control group, EGF combined AG1478 group and AG1478 group ( P<0.05) . Those in IL-8 group were higher than that in control group, IL-8I group, IL-8 combined IL-8I group. Those in EGF combined IL-8 group was higher than that in IL-8 group and EGF group (P<0.05). Conclusion: IL-8 and EGF can pro-mote the proliferation of melanoma cells. There may be a synergistic effect between IL-8 and EGF, which may be related to the Akt and MAPK pathway.
