目的:观察通心络胶囊对心肌缺血大鼠血管新生功能的影响并探讨其作用机制。方法60只 SD 大鼠随机分为正常组、模型组、通心络低剂量组、通心络高剂量组、bFGF 阳性药对照组,结扎冠状动脉左前降支制作大鼠心肌缺血模型,组织植块法培养心肌微血管内皮细胞(CMECs),观察在血管生成过程中通心络胶囊及各对照组含药血清对 CMECs“增殖”、“迁移”和“成管”不同环节的干预作用。实时定量聚合酶链反应(real - time PCR)检测各组血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)的表达水平。结果各药物组干预后,CMECs的增殖率、迁移率、成管率呈现出正常组> bFGF 组>通心络高剂量组>通心络低剂量组>模型组的趋势,通心络高剂量组对血管生成的不同环节均有明显促进作用并显著优于其他各组( P <0.01),与 bFGF 组比较无统计学差异(P >0.05)。通心络高剂量组 CMECs 中 VEGF、bFGF 表达上调,其中 VEGF 表达水平显著高于模型组和低剂量组(P <0.01)。结论通心络胶囊能够促进缺血 CMECs“增殖”、“迁移”和“成管”,发挥自身血管搭桥的作用,其机制可能与上调 VEGF、bFGF 表达有关。

목적:관찰통심락효낭대심기결혈대서혈관신생공능적영향병탐토기작용궤제。방법60지 SD 대서수궤분위정상조、모형조、통심락저제량조、통심락고제량조、bFGF 양성약대조조,결찰관상동맥좌전강지제작대서심기결혈모형,조직식괴법배양심기미혈관내피세포(CMECs),관찰재혈관생성과정중통심락효낭급각대조조함약혈청대 CMECs“증식”、“천이”화“성관”불동배절적간예작용。실시정량취합매련반응(real - time PCR)검측각조혈관내피세포생장인자(VEGF)、감성성섬유세포생장인자(bFGF)적표체수평。결과각약물조간예후,CMECs적증식솔、천이솔、성관솔정현출정상조> bFGF 조>통심락고제량조>통심락저제량조>모형조적추세,통심락고제량조대혈관생성적불동배절균유명현촉진작용병현저우우기타각조( P <0.01),여 bFGF 조비교무통계학차이(P >0.05)。통심락고제량조 CMECs 중 VEGF、bFGF 표체상조,기중 VEGF 표체수평현저고우모형조화저제량조(P <0.01)。결론통심락효낭능구촉진결혈 CMECs“증식”、“천이”화“성관”,발휘자신혈관탑교적작용,기궤제가능여상조 VEGF、bFGF 표체유관。
Objective:To investigate the effects of Tongxinluo capsule on the angiogenesis in rats of myocardial ischemia and its mechanisms. Methods:60 SD rats were divided into normal group,model group,low - dose group of Tongxinluo capsule,high - dose of group of Tongxinluo capsule and bFGF positive control group. We made the rat model of myocardial ischemia by ligate the anterior descending branch of coronary artery and culture the microvascular endothelial cells (CMECs)of myocardial muscles by explant method. We observe the intervention effects of Tongxinluo capsule and the con-trol medicated serums on proliferation,migration,angioblast of CMECs. Real - time PCR was used to detect the expression level of VEGF,bFGF in different groups. Results:After the drug intervention,the tendency of proliferation rate,migration rate and angioblast rate of CMECs from high to low were normal group,bFGF positive control group,high - dose of group of Tongxinluo capsule,low - dose group of Tongxinluo capsule,model group consequently. High - dose of Tongxinluo capsule could facilitate the angiogenesis in different aspects and better than other groups(p < 0. 01). Compared to the bFGF group, there was no statistically significance(P > 0. 05. The levels of VEGF and bFGF were upregulated of CMECs in group of high - dose Tongxinluo capsule. The expression levels of VEGF were higher than the model group and the low - dose Tongx-inluo capsule group(P < 0. 01). Conclusion:Tongxinluo capsule could facilitate the proliferation,migration and angioblast in ischemic CMECs. It might have the effect of vascular bypass which might related to the upregulation of MEGF and bF-GF.
