目的:观察姜黄素对胃癌组织中MMP-2和CD44v6在mRNA水平表达的影响,探讨其抑制胃癌细胞转移的作用机制。方法以SGC-7901胃癌细胞株建立胃癌裸鼠原位移植瘤模型,将裸鼠随机分为对照组及姜黄素组,每组12只。观察裸小鼠胃癌种植后肿瘤生长及转移灶情况,用原位杂交方法检测肿瘤组织中MMP-2 mRNA和CD44 v6 mRNA表达的变化。结果所有荷瘤鼠胃壁均有肿瘤生长,荷瘤率100%,姜黄素组肝、腹腔和淋巴结的转移比对照组明显减少( P <0ú.05);姜黄素治疗组瘤体中MMP-2 mRNA和CD44v6 mRNA的表达明显下调( P <0.05)。结论姜黄素具有抑制人胃癌裸鼠原位移植瘤转移的作用,其作用机制可能与下调MMP-2 mRNA和CD44 v6 mRNA的表达有关。

목적:관찰강황소대위암조직중MMP-2화CD44v6재mRNA수평표체적영향,탐토기억제위암세포전이적작용궤제。방법이SGC-7901위암세포주건립위암라서원위이식류모형,장라서수궤분위대조조급강황소조,매조12지。관찰라소서위암충식후종류생장급전이조정황,용원위잡교방법검측종류조직중MMP-2 mRNA화CD44 v6 mRNA표체적변화。결과소유하류서위벽균유종류생장,하류솔100%,강황소조간、복강화림파결적전이비대조조명현감소( P <0ú.05);강황소치료조류체중MMP-2 mRNA화CD44v6 mRNA적표체명현하조( P <0.05)。결론강황소구유억제인위암라서원위이식류전이적작용,기작용궤제가능여하조MMP-2 mRNA화CD44 v6 mRNA적표체유관。
Objective To investigate the effects of curcumin on the expression of MMP-2 mRNA and CD44v6 mRNA in human gastric cells ,and to explore the possible action mechanisms of curcumin in inhibiting metastasis of gastric cancer . Methods The orthotopic transplantation tumor models were established in nude mice with SGC-7901 gastric cancer cell line , then the nude mice were randomly divided into control group and curcumin group , with 12 mice in each group .The tumor growth and metastasis condition were observed ,and the expression levels of MMP-2 mRNA and CD44 v6 mRNA in tumor tissue were detected by in situ hybridization .Results The successful rate of orthotopic transplantation tumor was 100%.The metastases of liver, peritoneal cavity and lymph node in curcumin group were significantly less than those in control group (P<0.05),moreover, the expression levels of MMP-2 mRNA and CD44v6 mRNA in curcumin group were significantly down-regulated,as compared with those in control group ( P <0.05).Conclusion Curcumin can inhibit gastric cancer metastasis in nude mice orthotopic transplantation tumor ,and its action mechanism might be related to down-regulation of the expression levels of MMP-2 mRNA and CD44v 6 mRNA.
