目的::观察阿托伐他汀对心肌梗死大鼠心室重构和 miRNA-21表达的影响。方法:将48只健康 Wistar大鼠随机分为假手术组、模型组、阿托伐他汀低剂量治疗组和高剂量治疗组,后3组结扎左冠状动脉前降支建立急性心肌梗死(AMI)模型,阿托伐他汀低剂量和高剂量治疗组分别按20和40 mg/(kg·d)的剂量灌胃给药。6周后测定左室质量指数(LVMI)、心肌胶原容积分数(CVF),RT-PCR 法检测梗死周边区心肌组织 miRNA-21及 MMP-9 mRNA 的表达。结果:4组大鼠 LVMI、心肌 CVF 及梗死周边区心肌组织中 miRNA-21和 MMP-9 mRNA 的表达水平差异均有统计学意义(F =53.021、54.003、42.008和45.002,P 均<0.05)。与模型组相比,阿托伐他汀治疗组上述指标均降低(P <0.05),低剂量和高剂量治疗组间上述指标的差异无统计学意义(P >0.05)。模型组、阿托伐他汀低剂量和高剂量治疗组梗死周边区心肌组织 miRNA-21表达水平与 MMP-9 mRNA 水平呈正相关(r =0.613、0.512、0.551,P <0.05)。结论:阿托伐他汀能降低梗死周边区心肌组织 MMP-9 mRNA 和 miRNA-21的表达,从而发挥缩小梗死面积、减轻心室重构的作用。

목적::관찰아탁벌타정대심기경사대서심실중구화 miRNA-21표체적영향。방법:장48지건강 Wistar대서수궤분위가수술조、모형조、아탁벌타정저제량치료조화고제량치료조,후3조결찰좌관상동맥전강지건립급성심기경사(AMI)모형,아탁벌타정저제량화고제량치료조분별안20화40 mg/(kg·d)적제량관위급약。6주후측정좌실질량지수(LVMI)、심기효원용적분수(CVF),RT-PCR 법검측경사주변구심기조직 miRNA-21급 MMP-9 mRNA 적표체。결과:4조대서 LVMI、심기 CVF 급경사주변구심기조직중 miRNA-21화 MMP-9 mRNA 적표체수평차이균유통계학의의(F =53.021、54.003、42.008화45.002,P 균<0.05)。여모형조상비,아탁벌타정치료조상술지표균강저(P <0.05),저제량화고제량치료조간상술지표적차이무통계학의의(P >0.05)。모형조、아탁벌타정저제량화고제량치료조경사주변구심기조직 miRNA-21표체수평여 MMP-9 mRNA 수평정정상관(r =0.613、0.512、0.551,P <0.05)。결론:아탁벌타정능강저경사주변구심기조직 MMP-9 mRNA 화 miRNA-21적표체,종이발휘축소경사면적、감경심실중구적작용。
Aim: To explore the effect of atorvastatin on ventricular remodeling and expression of miRNA-21 in rats with myocardial infarction. Methods: A total of 48 male Wistar rats were randomly allocated into 4 groups:sham operation group, model group, low-dose atorvastatin and high-dose atorvastatin group,12 rats in each group. The rats in the latter 3 groups were made the acute myocardial infarction(AMI) model by ligating the left coronary artery. Low-dose atorvastatin and high-dose atorvastatin groups were given atorvastatin at the doses of 20 and 40 mg/ (kg·d) for 6 weeks,respectively. Then,left ventricular mass index(LVMI),collagen volume fraction(CVF),and the expressions of MMP-9 mRNA and miR-NA-21 in the surrounding infarct areas were detected. Results: There were significant differences in LVMI, CVF, the ex-pressions of miRNA-21 and MMP-9 mRNA among the four groups(F = 53. 021,54. 003,42. 008 and 45. 002,P < 0. 05). LVMI, CVF,and the expressions of miRNA-21 and MMP-9 mRNA were significantly decreased in atorvastatin groups com-pared with those of the model group(P < 0. 05),while no significant differences were found between low-dose and high-dose atorvastatin groups(P > 0. 05). The expression of miRNA-21 was positively correlated with MMP-9 mRNA in the model, low-dose and high-dose atorvastatin group(r = 0. 613,0. 512,0. 551,P < 0. 05). Conclusion: Atorvastatin could decrease the expressions of MMP-9 mRNA and miRNA-21 in rats with AMI, which could decrease infarct area and improve ventricu-lar remodeling and heart function.
