目的 研究表明支气管哮喘(简称哮喘)患者血清中IL-25的含量显著增加,并且与气道重塑高度相关,然而其具体机制尚未见报道.本文旨在探讨IL-25对大鼠气道平滑肌细胞增殖活性的影响及其机制.方法 原代培养大鼠气道平滑肌细胞,IL-25处理后,采用MTT法检测细胞增殖活性,并运用流式细胞术检测细胞周期.进一步采用real-time PCR法和western blot法检测细胞周期蛋白eyclinD1 mRNA和蛋白表达,探讨IL 25对气道平滑肌细胞增殖活性影响的机制.结果 MTT结果显示IL-25可呈剂量和浓度依赖性地上调原代大鼠气道平滑肌细胞的增殖活性,流式细胞术检测结果表明IL-25可促进细胞G1期向S期转变,其可能机制与IL-25上调细胞周期蛋白cyclin D1的表达有关.结论 IL-25可能是哮喘时导致气道重塑的重要因素之一.

Objective The level of interleukin-25 (IL-25) in serum of patients with bronchial asthma (asthma) was reported significantly increasing,and highly correlated with airway remodeling.But the exact mechanism has not been reported.This study aims to investigate the effect of IL-25 on proliferation of airway smooth muscle cell in rats.Methods Primary rat airway smooth muscle cells were treated with IL-25.MTT assay was used to detect cell proliferation and cell cycle was detected by flow cytometry.Real-time PCR and Western blot were employed to detect the mRNA and protein expression of cyclinD1,to explore the mechanism of IL-25 on cell proliferation of airway smooth muscle.Results MTT results showed that IL-25 increased the cell proliferation of primary rat airway smooth muscle cell in a dose-and concentration-dependent manner.Flow cytometry results showed that IL-25 can promote cell G1 to S phase transition,which mechanism and IL-25 may regulate the expression of cell cycle related proteins cyclin D1.Conclusions IL-25 may be an important factor leading to airway remodeling in asthma.
