目的:探讨三黄糖肾康对2型糖尿病并发骨质疏松症大鼠瘦素表达的影响。方法2013年6月—2014年11月取雌性Wistar大鼠,采用随机数字表法分为正常对照组10只,骨质疏松症模型组10只和糖尿病模型组50只。糖尿病模型组大鼠进一步按血糖水平随机分为糖尿病模型对照组、中药预防组、中药高剂量组、中药低剂量组和西药组(每组10只)。7组大鼠分别给予相应干预,喂养20周后取材检测大鼠空腹血糖、血清瘦素、骨密度、骨组织瘦素mRNA和瘦素表达水平、下丘脑瘦素长型受体( LRb)、磷酸化信号转导与转录激活因子3( p-STAT3)和磷酸化信号转导与转录激活因子5(p-STAT5)的表达水平。结果中药预防组、中药高剂量组、中药低剂量组和西药组大鼠空腹血糖水平、骨组织瘦素mRNA和瘦素表达水平、下丘脑LRb表达水平较正常对照组和骨质疏松症模型组升高,较糖尿病模型对照组下降( P<0.05);血清瘦素水平、下丘脑p-STAT3和p-STAT5表达水平较正常对照组升高,较骨质疏松症模型组和糖尿病模型对照组下降(P<0.05);骨密度较骨质疏松症模型组升高(P<0.05)。结论三黄糖肾康可能通过改善瘦素抵抗,直接或间接调节瘦素对骨代谢的影响,从而对2型糖尿病并发骨质疏松症起到治疗作用。
目的:探討三黃糖腎康對2型糖尿病併髮骨質疏鬆癥大鼠瘦素錶達的影響。方法2013年6月—2014年11月取雌性Wistar大鼠,採用隨機數字錶法分為正常對照組10隻,骨質疏鬆癥模型組10隻和糖尿病模型組50隻。糖尿病模型組大鼠進一步按血糖水平隨機分為糖尿病模型對照組、中藥預防組、中藥高劑量組、中藥低劑量組和西藥組(每組10隻)。7組大鼠分彆給予相應榦預,餵養20週後取材檢測大鼠空腹血糖、血清瘦素、骨密度、骨組織瘦素mRNA和瘦素錶達水平、下丘腦瘦素長型受體( LRb)、燐痠化信號轉導與轉錄激活因子3( p-STAT3)和燐痠化信號轉導與轉錄激活因子5(p-STAT5)的錶達水平。結果中藥預防組、中藥高劑量組、中藥低劑量組和西藥組大鼠空腹血糖水平、骨組織瘦素mRNA和瘦素錶達水平、下丘腦LRb錶達水平較正常對照組和骨質疏鬆癥模型組升高,較糖尿病模型對照組下降( P<0.05);血清瘦素水平、下丘腦p-STAT3和p-STAT5錶達水平較正常對照組升高,較骨質疏鬆癥模型組和糖尿病模型對照組下降(P<0.05);骨密度較骨質疏鬆癥模型組升高(P<0.05)。結論三黃糖腎康可能通過改善瘦素牴抗,直接或間接調節瘦素對骨代謝的影響,從而對2型糖尿病併髮骨質疏鬆癥起到治療作用。
목적:탐토삼황당신강대2형당뇨병병발골질소송증대서수소표체적영향。방법2013년6월—2014년11월취자성Wistar대서,채용수궤수자표법분위정상대조조10지,골질소송증모형조10지화당뇨병모형조50지。당뇨병모형조대서진일보안혈당수평수궤분위당뇨병모형대조조、중약예방조、중약고제량조、중약저제량조화서약조(매조10지)。7조대서분별급여상응간예,위양20주후취재검측대서공복혈당、혈청수소、골밀도、골조직수소mRNA화수소표체수평、하구뇌수소장형수체( LRb)、린산화신호전도여전록격활인자3( p-STAT3)화린산화신호전도여전록격활인자5(p-STAT5)적표체수평。결과중약예방조、중약고제량조、중약저제량조화서약조대서공복혈당수평、골조직수소mRNA화수소표체수평、하구뇌LRb표체수평교정상대조조화골질소송증모형조승고,교당뇨병모형대조조하강( P<0.05);혈청수소수평、하구뇌p-STAT3화p-STAT5표체수평교정상대조조승고,교골질소송증모형조화당뇨병모형대조조하강(P<0.05);골밀도교골질소송증모형조승고(P<0.05)。결론삼황당신강가능통과개선수소저항,직접혹간접조절수소대골대사적영향,종이대2형당뇨병병발골질소송증기도치료작용。
Objective To explore the effects of Sanhuangtangshenkang on the expression of leptin in rats with type 2 diabetes mellitus complicated with osteoporosis .Methods Female Wistar rats were taken as the study subjects from June 2013 to November 2014.Assigned 10 rats into normal control group , 10 rats into osteoporosis model group and 50 rats into diabetes model group.The diabetes model group was further divided into diabetes model control group , TCM prevention group , high-dose TCM group, low-dose TCM group and western medicine group , with 10 rats each group.After the corresponding intervention for 20 weeks, samples were taken and tested to determine fasting blood glucose (FBG), the expression of serum leptin, bone density, the mRNA and expression of bone tissue leptin , the expression of leptin receptor ( LRb ) , p-STAT3 and p-STAT5 in hypothalamus.Results TCM prevention group , high -dose TCM group , low -dose TCM group and western medicine group were higher (P<0.05) than normal control group and osteoporosis model group and were lower (P<0.05) than diabetes model control group in FBG , the mRNA and expression of bone tissue leptin and the expression of LRb in hypothalamus ; TCM prevention group , high-dose TCM group , low -dose TCM group and western medicine group were higher ( P<0.05 ) than normal control group and were lower ( P<0.05) than osteoporosis model group and diabetes model control group in the expression level of serum leptin and the expression levels of p-STAT3 and p-STAT5 in hypothalamus;TCM prevention group , high-dose TCM group, low-dose TCM group and western medicine group were higher ( P<0.05) than osteoporosis model group in bone density.Conclusion Sanhuangtangshenkang can probably directly or indirectly regulate the effect of leptin on bone metabolism by improving leptin resistance , thus being effective in the treatment of type 2 diabetes mellitus complicated with osteoporosis rats .
