目的:研究川芎嗪(TMP)对脓毒症大鼠肝细胞线粒体结构的保护作用及可能机制。方法实验大鼠被分成对照组、脓毒症组、治疗组和预防组;用盲肠结扎穿孔法制备大鼠脓毒症实验模型,在术后10 h,取肝脏标本用 JEM-1230透射电子显微镜观察肝细胞的超微结构,选用 Image-Pro Plus 6.0图像软件对线粒体进行测量;用 Western blot 技术检测肝细胞线粒体内膜水通道蛋白(AQP)8的表达。结果和对照组比较,其余3组单个肝细胞切面的线粒体总数和数密度均减少(P <0.01),但治疗组和预防组均多于脓毒症组(P <0.01);线粒体体积密度脓毒症组也较对照组低(P <0.01);线粒体内膜 AQP8的相对表达量在脓毒症组、治疗组和预防组均较对照组明显减少(P <0.01);治疗组和预防组的 AQP8相对表达量较脓毒症组增多(P <0.01)。结论在脓毒症状态下,TMP 可保护肝细胞线粒体的结构,可能是通过增加线粒体内膜 AQP8表达而起作用的。

목적:연구천궁진(TMP)대농독증대서간세포선립체결구적보호작용급가능궤제。방법실험대서피분성대조조、농독증조、치료조화예방조;용맹장결찰천공법제비대서농독증실험모형,재술후10 h,취간장표본용 JEM-1230투사전자현미경관찰간세포적초미결구,선용 Image-Pro Plus 6.0도상연건대선립체진행측량;용 Western blot 기술검측간세포선립체내막수통도단백(AQP)8적표체。결과화대조조비교,기여3조단개간세포절면적선립체총수화수밀도균감소(P <0.01),단치료조화예방조균다우농독증조(P <0.01);선립체체적밀도농독증조야교대조조저(P <0.01);선립체내막 AQP8적상대표체량재농독증조、치료조화예방조균교대조조명현감소(P <0.01);치료조화예방조적 AQP8상대표체량교농독증조증다(P <0.01)。결론재농독증상태하,TMP 가보호간세포선립체적결구,가능시통과증가선립체내막 AQP8표체이기작용적。
Objective To study the protective effects and plausible mechanisms of tetramethylpyrazine (TMP)on the structure of hepatocyte mitochondria in septic rats.Methods The rats were randomly divided into four groups:control group,septic group,therapeutic group and prevention group.The model of septic rat was established by the cecal ligation and puncture(CLP).The liver was taken out from septic rat 10 h after CLP,.The ultrastructure of hepatocyte were observed by the JEM-1230 transmission electron microscope.The hepatocyte mitochondria were meas-ured by Image-Pro Plus 6.0 analysis software.The expression of AQP8 was assayed by Western blot.Results Com-pared with the control group,the total number of mitochondria and numberical density were fewer in other 3 groups. The total number of mitochondria and numberical density of septic group were more than the therapeutic and preven-tion group(P <0.01),but the volume density of septic group was lower than the control group(P <0.01).Compared with the control group,the relative AQP8 protein expression was significantly reduced in the other 3 groups(P <0.01).The AQP8 protein expression of the therapeutic and prevention group were higher than the septic group(P <0.01).Conclusion TMP can protect stereology of hepatocyte mitochondria in septicrats by increasing the expression of AQP8 protein.
