目的观察高湿环境下大鼠肝脏组织损伤及细胞凋亡情况,探讨高湿环境对大鼠肝脏的影响。方法32只SD大鼠随机分为两组,高湿20d组与对照组,每组16只大鼠。采用人工气候箱模拟高湿环境,条件设置为温度25℃、相对湿度(90%±2%)RH。高湿20d组大鼠每天置于气候箱中10h,其余时间与对照组饲养于相同环境,温度25℃,相对湿度(45%~50%)RH,时间为20d。动物模型制作完成后,取肝脏组织行免疫组化及Western实验观察两组肝脏细胞凋亡情况。结果高湿组Tunel法原位标记可观察到肝脏细胞凋亡,同时活化的Caspase-3酶表达显著升高。结论高湿环境可诱导大鼠肝脏细胞凋亡的发生,凋亡的关键酶Caspase-3在其中扮演了重要角色,为下一步研究高湿诱导细胞凋亡的具体机制提供了实验基础。

Objective To observe the damage of rat liver tissue and the apoptosis of rat liver cells in high humidity environment so as to discuss the influence of high humidity on rat liver. Methods 32 SD rats were randomly divided into 2 groups:high humidity group and control group, 16 in each; artificial climate box was applied in imitating the high humidity environment, with the temperature of 25℃and relative humidity of 90±2%RH;rats in both groups were fed in the same environment with the temperature of 25℃and rela-tive humidity of 45%-50%RH for 20 days, during which, rats in high humidity group were kept in the artificial climate box for 10 hours every day; immunohistochemical study and Western Bolt were applied in observing the apoptosis of liver cells in rats in both groups. Results Apoptosis of rat liver cells in high humidity group was observed by TUNEL and the expression of activated enzyme Caspase-3 was obviously high. Conclusions High humidity environment may induce the apoptosis of rat liver cells and Caspase-3 as the key enzyme plays an important role in the process, which provides an experimental basis for further research on the mechanism of apop-tosis induced in high humidity environment.
