采用双波长替代单波长,对考马斯亮蓝分光光度法测定蛋白质技术进行改进。结果表明:以酪蛋白为标准蛋白,选取600 nm和460 nm为测定波长和参比波长,建立了双波法标准曲线,工作曲线相关系数的平方R2=0.999,检测范围3~250μg/mL;干扰试验和重复实验说明该方法准确性高、稳定性好;利用双波长法测定牛奶样品中蛋白质含量,相对偏差小于2.720%。因此,双波长分光光度法是一种简便、快速、准确、实用的蛋白质含量测定方法。

To improve a method for determination protein content, dual-wavelength spectrophotometry was used instead of single wave?length spectrophotometry in this paper. The results showed that standard curve of double wavelength spectrophotometry was established for the standard protein casein when chosen wavelength of 600 nm and 460 nm. The standard curve was significant for the square of cor?relation coefficient R2=0.999 when protein content in the range of 3-250 μg/mL. It showed that this method was of high accuracy through interference test and repeat experiment. Applying dual wavelength method to assay protein content for fresh milk , the relative standard deviation was less than 2.72%. Therefore, the dual-wavelength spectrophotometry is a good and practical determination meth?od for protein content.
