对黄色蚕茧超声辅助酶法脱胶工艺进行研究。在单因素实验的基础上,选择对蚕茧脱胶率有显著影响的3个因素:p H (X1)、酶加量(X2)、超声功率(X3),进行三因素三水平的响应面分析实验。最终确定最优条件为:p H 5.5、酶加量5.5%(E/S)、超声功率390W、超声处理时间35min、温度60℃、料液比1∶30(m/v)。并且该最佳条件下脱胶率(37.62±0.34%)和脱下的丝胶蛋白DPPH自由基清除率(IC502.41±0.078 mg/mL )、类胡萝卜素含量(0.34±0.038 m g/g )和黄酮含量(0.59±0.062 m g/g )均高于水浴煮沸脱胶和酶法脱胶,进一步表明了超声辅助酶法脱胶的优越性。

The purpose of this research is to study the process of ultrasonic‐assisted enzymatic degumming of yellow cocoons .Based on the single factor tests ,the three factors :pH (X1 ) ,enzyme amount (X2 ) ,and the ratio of mate‐rial to solvent (X3 ) had a significant impact on degumming rate of natural yellow cocoons .Then the three factors were assessed by response surface methodology (RSM ) involving a three factors and three levels . The optimal process was determined as follows:the pH was 5 .5 ,the enzyme amount was 5 .5% (E/S) ,the ultrasonic power was 390 W ,the ultrasonic treatment time was 35 min ,the temperature was 60℃ ,and the ratio of material to sol‐vent was 1 ∶ 30(m/v) .Under such conditions ,the ultrasonic‐assisted enzymatic has higher degumming rate of 37 .62% ± 0 .34% ,and the sericin protein after ultrasonic‐assisted enzymatic degumming has higher DPPH scaven‐ging activity (IC50 2 .41 ± 0 .078mg/mL ) ,content of carotenoids (0 .34 ± 0 .038mg/g ) ,and flavonoids (0 .59 ± 0 .062mg/g) than boiling water bath and enzymatic degumming .The ultrasonic‐assisted enzymatic degumming was further demonstrated superior .
