The optimal conditions of producing genistein demonstrated by applying sigle factor and response surface methodology were that the concentration of soybean meal, rotation speed, fermentation hours the optimal pH were 8 %, 180 r/min, 64 h, 7.3, the liquid loading amount was 50 mL/250 mL, the bacteria amount was 10 %. The yield of genistein ratio arrived at 0.12 %, the conversion rate was 89.13 %, which achieves the optimal level.
