目的:探讨SIRT1在有氧运动改善 APOE‐/‐小鼠肝脏脂代谢异常中的作用。方法:16只雄性 C57BL/6J小鼠随机分为 C57BL/6J小鼠对照组(CC)和运动组(CE),16只雄性APOE‐/‐小鼠随机分为APOE‐/‐小鼠对照组(AC )和运动组(AE ),每组8只。12周有氧运动干预后,通过病理学切片观察肝脏脂质积聚情况,检测小鼠肝脏 SIRT1、LKB1、P‐LKB1‐ser428、AMPKα1、P‐AMPKα1‐Thr172和 FAS蛋白表达量。结果:1)与 CC组比较,AC组肝脏脂肪积聚增加,与AC组比较,AE组肝脏脂质积聚减少;2)与CC组比较,CE组SIRT1( P<0.05)、P‐LKB1‐ser428(P<0.01)和 P‐AMPKα1‐Thr172(P<0.01)含量明显升高,LKB1、AMPKα1和FAS没有明显变化(P>0.05);3)与 CC 组比较,AC 组 SIRT1(P<0.05)、P‐LKB1‐ser428(P<0.01)和P‐AMPKα1‐Thr172(P<0.01)表达明显降低,LKB1、AMPKα1和FAS没有明显变化( P>0.05);4)与 AC组比较,AE组 SIRT 1( P<0.01)、P‐LKB1‐ser428(P<0.01)和P‐AMPKα1‐Thr172(P<0.01)表达明显升高,FAS表达明显降低(P<0.01), LKB1和AM PKα1没有明显变化( P>0.05)。结论:12周有氧运动可能通过增加SIRT 1表达,提高LKB1和 AMPKα1活性,抑制 FAS表达,改善APOE‐/‐小鼠肝脏脂代谢异常。

목적:탐토SIRT1재유양운동개선 APOE‐/‐소서간장지대사이상중적작용。방법:16지웅성 C57BL/6J소서수궤분위 C57BL/6J소서대조조(CC)화운동조(CE),16지웅성APOE‐/‐소서수궤분위APOE‐/‐소서대조조(AC )화운동조(AE ),매조8지。12주유양운동간예후,통과병이학절편관찰간장지질적취정황,검측소서간장 SIRT1、LKB1、P‐LKB1‐ser428、AMPKα1、P‐AMPKα1‐Thr172화 FAS단백표체량。결과:1)여 CC조비교,AC조간장지방적취증가,여AC조비교,AE조간장지질적취감소;2)여CC조비교,CE조SIRT1( P<0.05)、P‐LKB1‐ser428(P<0.01)화 P‐AMPKα1‐Thr172(P<0.01)함량명현승고,LKB1、AMPKα1화FAS몰유명현변화(P>0.05);3)여 CC 조비교,AC 조 SIRT1(P<0.05)、P‐LKB1‐ser428(P<0.01)화P‐AMPKα1‐Thr172(P<0.01)표체명현강저,LKB1、AMPKα1화FAS몰유명현변화( P>0.05);4)여 AC조비교,AE조 SIRT 1( P<0.01)、P‐LKB1‐ser428(P<0.01)화P‐AMPKα1‐Thr172(P<0.01)표체명현승고,FAS표체명현강저(P<0.01), LKB1화AM PKα1몰유명현변화( P>0.05)。결론:12주유양운동가능통과증가SIRT 1표체,제고LKB1화 AMPKα1활성,억제 FAS표체,개선APOE‐/‐소서간장지대사이상。
Objective :To observe the effect of SIRT1 of aerobic exercise on the improvement liver lipid metabolism in APOE‐/‐ rats .Methods :16 male C57BL/6J rats were randomly divid‐ed into control group (CC ,n= 8) ,exercise group (CE ,n= 8) ,16 male APOE‐/‐ rats were ran‐domly divided into control group (AC ,n=8) ,exercise group (AE ,n=8) .After 12 weeks in‐tervention ,hepatic lipid accumulation were observed ,Also the protein expression of SIRT1 , LKB1 ,P‐LKB1‐ser428 ,AMPKα1 ,P‐AMPKα1‐Thr172 and FAS were determined in the liv‐er .Results :1) Compared with CC group ,hepatic lipid accumulation increased significantly in the AC group ,Compared with AC ,lipid accumulation decreased significantly in the AE group . 2) Compared with CC group ,the content of SIRT1 (P<0 .05) ,P‐LKB1‐ser428(P< 0 .01) and P‐AMPKα1‐Thr172(P< 0 .01)increased significantly ,while the expression of LKB1 , AMPKα1 and FAS (P> 0 .05)didn’t change significantly in the CE group .3) Compared with CC group ,the expression of SIRT 1 ( P < 0 .05 ) ,P‐LKB1‐ser‐428 ( P < 0 .01 ) and P‐AMPKα1‐Thr172 ( P < 0 .01 ) decreased significantly ,while the expression of LKB1 , AMPKα1and FAS(P> 0 .05)didn’t change significantly in the liver of AC group .4)Com‐pared with AC group ,the content of SIRT1 (P< 0 .01) ,P‐LKB1‐ser428(P< 0 .01)and P‐AMPKα1‐Thr172(P<0 .01)increased significantly ,while that of FAS(P< 0 .01)decreased significantly ,and that of LKB1 and AM PKα1 didn’ t change obviously ( P> 0 .05 ) .Conclu‐sions :12 weeks aerobic exercise may be through promoting the SIRT1 ,increasing LKB1 and AM PKα1 activity ,inhibiting the expression of FAS ,to ameliorate hepatic lipid metabolism dis‐order of APOE‐/‐ rats .
