为给水生植物芡实(Euryale ferox)生物肥料的研制提供优良的菌株资源,采用保绿法和萝卜子叶增重法从芡实根际土壤筛选出2株作用效果较好的细菌 Q6-2和 Q67,用高效液相色谱法(High Performance Liquid Chromatography,HPLC)定量分析菌株产植物激素和有机酸的能力;通过盆栽试验研究2株细菌对苗期芡实的促生作用;结合生理生化试验和16S rDNA 序列分析对2株细菌进行鉴定。结果显示,2株细菌均能够产生玉米素、激动素等植物激素和少量有机酸,Q6-2还能够产生吲哚乙酸;接种 Q6-2后,芡实幼苗茎长、叶片数、最大叶面积、生物量(干重)分别比对照高出18.85%、40.67%、76.28%和37.50%,接种 Q67的处理各项指标分别比对照高出27.47%、42.81%、96.05%和42.97%,差异显著;接种 Q6-2和 Q67后,叶绿素含量分别高出对照43.27%和51.46%,根系活力则分别高出5.41%和7.05%,差异显著。经鉴定,Q6-2和 Q67均为地衣芽孢杆菌,GenBank 登录号分别为 KP686129和 KP686133。2株细菌有效促进了芡实幼苗生长发育水平,在微生物肥料的生产中具有广阔的应用前景。

This study aims to provide excellent bacterial strain resources for the production of Euryale fer-ox bio -fertilizer.Two strains with relatively higher performance named Q6 -2 and Q67 were screened from the rhizosphere soil of Euryale ferox using the methods of remaining green and radish cotyledon bioassay;their production capability of plant hormones and organic acids were quantitatively analyzed by HPLC;their promo-tion effects on the growth of Euryale ferox at seedling stage were investigated by pot experiments;also,they were identified by physiological and biochemical experiments and 16S rDNA sequence analysis.The results in-dicated that the two strains were both able to produce zeatin,kinetin and small amount of organic acid.Q6 -2 was also able to produce indoleacetic acid (IAA).After inoculated by Q6 -2,the growth parameters of Eury-ale ferox seedlings,such as stem length,number of leaves,maximum leaf area and biomass (dry weight), significantly increased by 18.85%,40.67%,76.28% and 37.50% respectively compared to the control;while inoculated by Q67,the above mentioned indexes significantly increased by 27.47%,42.81%, 96.05% and 42.97% respectively.After inoculated by Q6 -2 and Q67,the chlorophyll content increased by 43.27% and 51.46% respectively,and the root activity increased by 5.41% and 7.05% respectively,show-ing significant differences compared to the control.Q6 -2 and Q67 were both identified as Bacillus lichenifor-mis,whose GenBank accession number were KP686129 and KP686133 respectively.In conclusion,the two strains were able to promote the growth and development of Euryale ferox seedlings efficiently,showing great application prospect in the production of microbial fertilizer.
