目的:探讨环境毒物多环芳烃(polycyclic aromatic hydrocarbons,PAHs)对体外培养的卵巢组织中卵泡及其凋亡的影响。方法选取育龄期雌性小鼠卵巢,随机分为两组:对照组和 PAH 暴露组(0.1μg/ml、1μg/ml、10μg/ml),体外培养10天后进行组织切片、HE 染色观察卵巢形态和闭锁卵泡计数。采用免疫组化方法、Western blot 检测凋亡相关的 Caspase 3、Caspase 9、Bcl2/Bax。结果 PAHs 暴露组卵巢中卵泡数目减少,明显低于对照组(P <0.05),与对照组相比,PAH 组卵巢相关凋亡蛋白Caspase 3、Caspases 9、Bax 表达增加,抑制凋亡的 Bcl-2表达下降。结论育龄期雌鼠接触 PAHs 会导致卵泡数量的减少,其可能的机制是通过诱导卵巢颗粒细胞凋亡。

목적:탐토배경독물다배방경(polycyclic aromatic hydrocarbons,PAHs)대체외배양적란소조직중란포급기조망적영향。방법선취육령기자성소서란소,수궤분위량조:대조조화 PAH 폭로조(0.1μg/ml、1μg/ml、10μg/ml),체외배양10천후진행조직절편、HE 염색관찰란소형태화폐쇄란포계수。채용면역조화방법、Western blot 검측조망상관적 Caspase 3、Caspase 9、Bcl2/Bax。결과 PAHs 폭로조란소중란포수목감소,명현저우대조조(P <0.05),여대조조상비,PAH 조란소상관조망단백Caspase 3、Caspases 9、Bax 표체증가,억제조망적 Bcl-2표체하강。결론육령기자서접촉 PAHs 회도치란포수량적감소,기가능적궤제시통과유도란소과립세포조망。
Objective To explore the effect of the environmental toxicants polycyclic aromatic hydrocarbons (PAHs) on apoptosis of follides in the ovary in vitro. Method The child-bearing period female mice were randomly divided into two groups: control group and PAHs exposure group (0.1 μg/ml, 1 μg/ml, 10 μg/mL).We took out their ovaries under aseptic conditions then culured the ovaries in vitro for 10 days . The structural changes of the ovaries were observed by the light microscope using HE staining .The apoptosis of the granular cells were detected by immunohistochemistry and the atresia follicles were counted . Results The follicle number of the PAHs exposed group was significantly lower than the control group (P < 0.05). Compared with the control group, the expressions of apoptosis protein Caspase 3, Caspases 9 and Bax were increased, the expression of Bcl-2 was decreased.Conclusions Females of reproductive age contacting PAHs will lead to a decrease in the number of ovarian.
