为鉴定猪传染性胃肠炎病毒（TGEV）N蛋白在猪小肠黏膜上皮细胞（SIEC）中的表达并检测其对细胞周期的影响，将构建的含有 TGEV N 基因的重组质粒 pEGFP-N1-N 采用脂质体介导法瞬时转染SIEC，通过 RT-PCR 和 Western blot 方法分别从分子水平和蛋白水平检测 N 蛋白的表达，采用流式细胞术检测 N 蛋白对 IEC 细胞周期的影响。结果表明，N 蛋白在 SIEC 中成功表达，并使细胞的 S 期延长。研究结果为深入研究 TGEV N 蛋白功能提供新的理论依据，并为进一步探讨 TGEV 的致病机理奠定基础。

To identify the expression of the N gene of TGEV in porcine swine intestinal epithelial cells (SIEC)and the effect of N gene on cell cycle,recombinant plasmids pEGFP-N1-N were transfected into SIEC using lipofectamine 2000.RT-PCR and Western blot were used to detect the expression of N gene through the level of molecule and protein respectively.The effect of N protein on cell cycle progression was investigated by flow cytometry assay.The results showed that the N gene of TGEV was successfully expressed in SIEC,resulting in the prolong of S-phase cell cycle.This research may contribute to establish the theoretical basis for further studying the function and the pathogenesis mechanism of N protein of TGEV.
