为鉴定猪传染性胃肠炎病毒(TGEV)N蛋白在猪小肠黏膜上皮细胞(SIEC)中的表达并检测其对细胞周期的影响,将构建的含有 TGEV N 基因的重组质粒 pEGFP-N1-N 采用脂质体介导法瞬时转染SIEC,通过 RT-PCR 和 Western blot 方法分别从分子水平和蛋白水平检测 N 蛋白的表达,采用流式细胞术检测 N 蛋白对 IEC 细胞周期的影响。结果表明,N 蛋白在 SIEC 中成功表达,并使细胞的 S 期延长。研究结果为深入研究 TGEV N 蛋白功能提供新的理论依据,并为进一步探讨 TGEV 的致病机理奠定基础。
為鑒定豬傳染性胃腸炎病毒(TGEV)N蛋白在豬小腸黏膜上皮細胞(SIEC)中的錶達併檢測其對細胞週期的影響,將構建的含有 TGEV N 基因的重組質粒 pEGFP-N1-N 採用脂質體介導法瞬時轉染SIEC,通過 RT-PCR 和 Western blot 方法分彆從分子水平和蛋白水平檢測 N 蛋白的錶達,採用流式細胞術檢測 N 蛋白對 IEC 細胞週期的影響。結果錶明,N 蛋白在 SIEC 中成功錶達,併使細胞的 S 期延長。研究結果為深入研究 TGEV N 蛋白功能提供新的理論依據,併為進一步探討 TGEV 的緻病機理奠定基礎。

To identify the expression of the N gene of TGEV in porcine swine intestinal epithelial cells (SIEC)and the effect of N gene on cell cycle,recombinant plasmids pEGFP-N1-N were transfected into SIEC using lipofectamine 2000.RT-PCR and Western blot were used to detect the expression of N gene through the level of molecule and protein respectively.The effect of N protein on cell cycle progression was investigated by flow cytometry assay.The results showed that the N gene of TGEV was successfully expressed in SIEC,resulting in the prolong of S-phase cell cycle.This research may contribute to establish the theoretical basis for further studying the function and the pathogenesis mechanism of N protein of TGEV.
