目的：通过观察白藜芦醇对结肠癌细胞系 HT -29增殖的影响并检测程序性细胞死亡4蛋白表达水平，探讨白藜芦醇的抗结肠癌增殖作用及部分相关机制。方法 HT -29细胞经不同浓度白藜芦醇作用后，流式细胞术检测细胞周期分布及凋亡情况，Western blot 法检测程序性细胞死亡4蛋白表达水平变化。结果不同浓度的白藜芦醇对 HT -29细胞的增殖有抑制作用，并且呈现出一定的剂量依赖性（ P <0.01），白藜芦醇改变细胞周期分布，细胞周期阻滞在 S 期，同时诱导 HT -29细胞凋亡，试验组与空白对照组相比，S 期细胞比例及凋亡率明显增高（P <0.05），细胞周期被阻滞，增殖抑制。白藜芦醇处理后 HT -29细胞程序性细胞死亡4蛋白表达水平上调，与对照组比较，差异有统计学意义（P <0.01）。结论白藜芦醇能够显著抑制 HT -29细胞株的增殖，细胞周期阻滞于 S 期，诱导肿瘤细胞凋亡，其肿瘤抑制作用可能与程序性细胞死亡4蛋白表达上调有关。
목적：통과관찰백려호순대결장암세포계 HT -29증식적영향병검측정서성세포사망4단백표체수평，탐토백려호순적항결장암증식작용급부분상관궤제。방법 HT -29세포경불동농도백려호순작용후，류식세포술검측세포주기분포급조망정황，Western blot 법검측정서성세포사망4단백표체수평변화。결과불동농도적백려호순대 HT -29세포적증식유억제작용，병차정현출일정적제량의뢰성（ P <0.01），백려호순개변세포주기분포，세포주기조체재 S 기，동시유도 HT -29세포조망，시험조여공백대조조상비，S 기세포비례급조망솔명현증고（P <0.05），세포주기피조체，증식억제。백려호순처리후 HT -29세포정서성세포사망4단백표체수평상조，여대조조비교，차이유통계학의의（P <0.01）。결론백려호순능구현저억제 HT -29세포주적증식，세포주기조체우 S 기，유도종류세포조망，기종류억제작용가능여정서성세포사망4단백표체상조유관。
Objective To evaluate the effect and mechanism of resveratrol inhibiting proliferation and inducing apopto-sis on human colon cancer cells HT - 29. Methods The proliferation of human colon cancer line HT - 29 was measured by CCK - 8 assay. Cell cycle and apoptosis were measured using Flow cytometer(FCM). Western blotting was used to detect PDCD4 protein expression. Results Different concentrations of resveratrol on cell growth was inhibited in a dose - depend-ent manner. The effect on cycle distribution and appotosis were significance difference(P < 0. 05). There was significant difference of the expression of PDCD4 on HT - 29 cells before it treated by resveratrol and after that(P < 0. 01). Conclu-sion Resveratrol could inhibite the proliferation,and induce cell cycle arrest and appotosis of HT - 29 cell line,which may be related to overexpression of PDCD4 on HT - 29 cells.
