目的:研究胃癌组织中音猬因子(Shh)、Patched1(Ptch1)基因的表达及意义。方法:采用反转录-聚合酶链反应(RT -PCR)和 Westem blot 检测98例胃癌组织和配对的癌旁组织中 Shh、Ptch 1 mRNA 和蛋白的表达情况。结果:RT -PCR 检测结果显示,Shh、Ptch 1 mRNA 在胃癌中相对表达量分别为0.687±0.057、0.594±0.046,在胃癌旁组织中分别为0.314±0.025、0.293±0.074(P <0.05)。Western blot 检测结果显示,Shh、Ptch1蛋白在胃癌中的相对表达量分别为0.525±0.057、0.481±0.046,在胃癌旁组织中分别为0.236±0.025、0.215±0.074。胃癌组织 Shh、Ptch1 mRNA 和蛋白平均表达水平高于癌旁组织,差异有统计学意义(P <0.05);Shh、Ptch1 mRNA 和蛋白表达水平与胃癌的分化程度、TNM分期、浸润深度和淋巴结转移明显相关(P <0.05),与患者年龄、性别和肿瘤直径无明显相关(P >0.05)。结论:胃癌组织中 Shh、Ptch1蛋白呈高表达,Shh、Ptch1蛋白的高表达可能与胃癌的发生及发展有关。

목적:연구위암조직중음위인자(Shh)、Patched1(Ptch1)기인적표체급의의。방법:채용반전록-취합매련반응(RT -PCR)화 Westem blot 검측98례위암조직화배대적암방조직중 Shh、Ptch 1 mRNA 화단백적표체정황。결과:RT -PCR 검측결과현시,Shh、Ptch 1 mRNA 재위암중상대표체량분별위0.687±0.057、0.594±0.046,재위암방조직중분별위0.314±0.025、0.293±0.074(P <0.05)。Western blot 검측결과현시,Shh、Ptch1단백재위암중적상대표체량분별위0.525±0.057、0.481±0.046,재위암방조직중분별위0.236±0.025、0.215±0.074。위암조직 Shh、Ptch1 mRNA 화단백평균표체수평고우암방조직,차이유통계학의의(P <0.05);Shh、Ptch1 mRNA 화단백표체수평여위암적분화정도、TNM분기、침윤심도화림파결전이명현상관(P <0.05),여환자년령、성별화종류직경무명현상관(P >0.05)。결론:위암조직중 Shh、Ptch1단백정고표체,Shh、Ptch1단백적고표체가능여위암적발생급발전유관。
Objective:To explore the expression and significance of sonic hedgehog(Shh),Patched1 (Ptch1)in human gastric cancer tissue.Methods:The expression of Shh,Ptch1 mRNA and protein was detected in 9 8 human gastric cancer tissue by RT -PCR and Western blot.Results:The expression of Shh,Ptch 1 mRNA in gastric cancer was 0.687 ±0.057,0.594 ±0.046,and in gastric cancer adjacent tissue was 0.314 ±0.025,0.293 ±0.074 (P <0.05).The expression of Shh,Ptch1 protein in gastric cancer was 0.525 ±0.057,0.481 ±0.046,and in gastric cancer adjacent tissue was 0.236 ±0.025,0.215 ±0.074 (P <0.05).The expression of Shh,Ptch1 mRNA and pro-tein in gastric cancer tissue were both higher than gastric cancer adjacent tissue.The expression of Shh,Ptch1 was sig-nificant associated with differentitation,TNMgrade,depth of invasion and lymph node metastasis(P <0.05),not age, gender and tumorous size (P >0.05).Conclusion:Shh,Ptch1 overexpressed in gastric cancer tissues and its abnor-mal expression may be involved in the oncogenesis and development of gastric cancer.
