目的:研究当归补血汤对高糖条件下大鼠肾小球系膜细胞( GMCs )核因子相关因子2(Nrf2)表达及总超氧化物歧化酶(T-SOD)活性、丙二醛(MDA )含量的影响,探讨其在糖尿病肾病( DN)治疗中的作用机制。方法将60只大鼠分为正常组、高糖组、当归补血汤高剂量组[7 g/(kg? d)]、当归补血汤中剂量组[3.5 g/(kg? d)]、当归补血汤低剂量组[1.75 g/(kg? d)]、格列奇特组[(2 mg/kg? d)],每组10只。应用RT-PCR法检测各组GMCs的Nrf2 mRNA表达情况,Western blot法检测各组GMCs的Nrf2蛋白表达情况,应用黄嘌呤氧化酶法检测各组GMCs培养上清液的T-SOD活性,应用硫代巴比妥酸法检测各组GMCs 培养上清液的MDA含量。结果正常组Nrf2 mRNA和蛋白有一定量的表达,高糖组和正常组相比Nrf2 mRNA和蛋白表达均增加,当归补血汤高剂量组对高糖条件下系膜细胞Nrf2 mRNA和蛋白表达均有明显上调作用(P均<0.01);与正常组相比,高糖组T-SOD 活性明显降低(P<0.01),MDA含量明显增多(P<0.01),而当归补血汤各剂量组能提高T-SOD的活性,抑制高糖条件下MDA的过多生成,尤其以高剂量组最为明显(P<0.01)。结论当归补血汤能上调高糖条件下系膜细胞Nr2f 的表达,提高T-SOD的活性,抑制MDA的过多生成,从而抵抗高糖诱导的氧化应激反应,保护系膜细胞。

목적:연구당귀보혈탕대고당조건하대서신소구계막세포( GMCs )핵인자상관인자2(Nrf2)표체급총초양화물기화매(T-SOD)활성、병이철(MDA )함량적영향,탐토기재당뇨병신병( DN)치료중적작용궤제。방법장60지대서분위정상조、고당조、당귀보혈탕고제량조[7 g/(kg? d)]、당귀보혈탕중제량조[3.5 g/(kg? d)]、당귀보혈탕저제량조[1.75 g/(kg? d)]、격렬기특조[(2 mg/kg? d)],매조10지。응용RT-PCR법검측각조GMCs적Nrf2 mRNA표체정황,Western blot법검측각조GMCs적Nrf2단백표체정황,응용황표령양화매법검측각조GMCs배양상청액적T-SOD활성,응용류대파비타산법검측각조GMCs 배양상청액적MDA함량。결과정상조Nrf2 mRNA화단백유일정량적표체,고당조화정상조상비Nrf2 mRNA화단백표체균증가,당귀보혈탕고제량조대고당조건하계막세포Nrf2 mRNA화단백표체균유명현상조작용(P균<0.01);여정상조상비,고당조T-SOD 활성명현강저(P<0.01),MDA함량명현증다(P<0.01),이당귀보혈탕각제량조능제고T-SOD적활성,억제고당조건하MDA적과다생성,우기이고제량조최위명현(P<0.01)。결론당귀보혈탕능상조고당조건하계막세포Nr2f 적표체,제고T-SOD적활성,억제MDA적과다생성,종이저항고당유도적양화응격반응,보호계막세포。
Objective It is to observe the effects of Danggui Buxue decoction on glomerular mesangial cell Nrf 2 expression and T-SOD activity , MDA content in high glucose condition ,and to explore its treatment significance in the process of diabet-ic nephropathy (DN).Methods Sixty rats were randomly divided into normal group , high glucose group, high, medium and low dose of Danggui Buxue decoction groups [7 g/(kg? d), 3.5 g/(kg? d),1.75 g/(kg? d)respectively], Gliclazide group[2 mg/(kg? d)], each group had 10 rats.RT-PCR was used to detect expression of Nrf2 mRNA and Western blot used to detect expression of Nrf 2 protein of GMCs in each group;Xanthine Oxidase assay was used to detect T -SOD activity and Thiobarbituric Acid method was used to detect MDA content in cell supernatant of GMCs in each group .Results Both of Nrf2 mRNA and protein in normal group had a certain amount of expression;Nrf2 mRNA (P<0.05) and protein (P<0.01) were both increased in high glucose group compared with normal group .High-dose group has a significantly upregulation effect on Nrf2 mRNA and protein expression of GMCs in high glucose condition (P<0.01).Compared with the normal group, T-SOD activity of high glucose group was significantly decreased (P<0.01), MDA content was significantly increased (P<0.01 );while SOD activity was improved , MDA production in high glucose condition was inhibited in every Dangui Buxue de -coction group, especially in the high dose group (P<0.01).Conclusion Danggui Buxue decoction could upregulate the ex-pression of Nrf2 mRNA and protein,improve SOD activity,inhibit MDA excessive generation of GMCs in high glucose condi-tion, thereby resisting oxidative stress induced by high glucose and protecting mesangial cells .
