目的 验证在小鼠下肢急慢性动脉缺血模型(部分/完全阻塞股动脉)中,外源性IMD是否能够促进下肢缺血组织血流恢复、增加新生血管标志物CD31、CD105标记的微血管密度.方法实验小鼠(C57BL/6)行股动脉完全阻断、动脉缝合部分阻断手术,并建立对照组.应用Doppler血流仪检测下肢血流评估缺血模型血流变化情况.术后立即对缺血下肢缝匠肌组织行外源性小鼠IMD多点注射,持续2 W后取缝匠肌组织标本.下肢缺血缝匠肌标本分别做HE病理染色及CD31、CD105免疫组织化学染色.结果 小鼠下肢动脉急慢性缺血模型建立后,下肢动脉血流供应阻断,Doppler血流提示下肢血流信号减少,术后随时间延长、IMD注射,缺血肢体血流信号有明显恢复,且IMD注射组与对照组及空白组相比血流恢复情况明显增加.下肢缺血缝匠肌组织病理HE染色显示,组织内微血管、血管周围单个或内皮细胞群数量IMD注射组较对照组多.CD31、CD105免疫组织化学染色结果显示IMD注射组微血管密度与对照组相比较差异有统计学意义(P<0.05).结论 小鼠急慢性下肢动脉缺血模型中,通过肌肉注射外源性IMD,能够促进缺血模型中下肢缺血组织的血流恢复和CD31、CD105免疫组化染色标记的新生血管内皮细胞密度增加.

목적 험증재소서하지급만성동맥결혈모형(부분/완전조새고동맥)중,외원성IMD시부능구촉진하지결혈조직혈류회복、증가신생혈관표지물CD31、CD105표기적미혈관밀도.방법실험소서(C57BL/6)행고동맥완전조단、동맥봉합부분조단수술,병건립대조조.응용Doppler혈류의검측하지혈류평고결혈모형혈류변화정황.술후립즉대결혈하지봉장기조직행외원성소서IMD다점주사,지속2 W후취봉장기조직표본.하지결혈봉장기표본분별주HE병리염색급CD31、CD105면역조직화학염색.결과 소서하지동맥급만성결혈모형건립후,하지동맥혈류공응조단,Doppler혈류제시하지혈류신호감소,술후수시간연장、IMD주사,결혈지체혈류신호유명현회복,차IMD주사조여대조조급공백조상비혈류회복정황명현증가.하지결혈봉장기조직병리HE염색현시,조직내미혈관、혈관주위단개혹내피세포군수량IMD주사조교대조조다.CD31、CD105면역조직화학염색결과현시IMD주사조미혈관밀도여대조조상비교차이유통계학의의(P<0.05).결론 소서급만성하지동맥결혈모형중,통과기육주사외원성IMD,능구촉진결혈모형중하지결혈조직적혈류회복화CD31、CD105면역조화염색표기적신생혈관내피세포밀도증가.
Objective The purpose of this study is to conform whether injected exogenous IMD can promote recovery of blood flow and angiogenesis on ischemic organization of the acute/chronic lower arterial ischemia model in mice (completely/partialy blocked femoral arteries).Methods Male mice (C57BL/6) femoral artery is completely blocked/partial suture blocked by surgery of acute/chronic lower arterial ischemia model.We use doppler imaging to assess the changes of blood flow.After the operation,we give exogenous IMD 150μg/kg body-weight for each mice by muscle injection, 1 time a day,lasts 2 weeks.Then we take ischemic lower limb skeletal muscle samples,which perform HE pathological staining and CD31,CD105 immuno-histochemical staining respectively, that can measure the density of neo-microvascular marked by CD31,CD105. Results The signal of lower limb blood flow on doppler blood flow imaging reduced when the models established.As time goes post-operation, ischemic limb blood flow signal substantialy restored, and recovery of blood flow in the IMD injection group is significantly higher than that in the control group. Pathologic HE staining of lower limb ischemia skeletal muscle showed that the number of single or grouped endothelial cels of blood vessels in IMD group is more than that in the control group.Results of CD31, CD105 immuno-histochemical stainings marked microvessel density in the IMD injection group was statisticaly significant difference (P<0.05) with the control group.Conclusion In the mice model of acute and chronic lower limb arterial ischemia, exogenous IMD can promote recovery of blood flow and microvessel angiogenesis, which conducive to the neovascularization of the ischemic tissue.
