建立高效液相色谱法同时测定半枝莲中原儿茶酸和香草酸含量的方法。采用Zorbax SB–C18柱(150 mm×4.6 mm,5μm),以0.2%磷酸水溶液–甲醇作淋洗液梯度洗脱,流量为1.0 mL/min,紫外检测波为260 nm,柱温为25℃。半枝莲中原儿茶酸和香草酸质量浓度分别在2.66~31.92μg/mL,2.86~34.32μg/mL与色谱峰面积呈良好的线性关系,线性相关系数r=0.9997,检出限分别为0.56,0.57μg/mL。原儿茶酸、香草酸的加样回收率分别为101.4%,99.4%,测定结果的相对标准偏差分别为1.76%,2.21%(n=9)。该方法适用于半枝莲中原儿茶酸和香草酸的含量测定。

A method for simultaneously determination of protocatechuic acid and vanillic acid in scutllaria barbata D.Don was established. A Zorbax SB–C18 column (250 mm×4.6 mm,5μm) was adopted with mobile phase of methanol–0.2%phosphoric acid with gradient elution at the flow rate of 1.0 mL/min,the detection wavelength was set at 260 nm and the column temperature was 25℃. The calibration curves for protocatechuic acid and vanillic acid were linear in the range of 2.66–31.92,2.86–34.32μg/mL,respectively. Both of the correlation coefficients were 0.999 7,the detection limits were 0.56, 0.57μg/mL,respectively. The average recoveries of protocatechuic acid and vanillic acid were 101.4%and 99.4%,respectively,the relative standard deviations were 1.76%and 2.21%(n=9). The method is suitable for determination of protocatechuic acid and vanillic acid in Scutllaria barbata D.Don.
