目的:观察不同性别和年龄健康人外周血单个核细胞(PBMC)中I型干扰素mRNA表达,探讨I型干扰素基因表达的性别和年龄差异。方法选择113例健康人,中老年54例包括女性27例和男性27例,平均(63±8)岁;青年人59例包括女性30例和男性29例,平均(36±6)岁。取抗凝全血,实时荧光定量PCR检测干扰素α(IFN-α)和干扰素β(IFN-β)的mRNA表达。结果健康中老年男性IFN-α和IFN-β基因表达量与青年男性比较,差异均有统计学意义(P<0.05),且前者基因表达量分别比后者低25倍和2.9倍;健康中老年女性IFN-α和IFN-β基因表达量与青年男性比较,差异均有统计学意义(P<0.05),且前者基因表达量分别比后者低6.67倍和3.57倍;健康青年男性IFN-α和IFN-β基因表达量与青年女性比较,差异均有统计学意义(P<0.05),且前者基因表达量分别比后者低4倍和1.92倍;健康中老年男性IFN-α基因表达量和中老年女性比较,差异有统计学意义(P<0.05),且前者基因表达量比后者低16.67倍,但健康中老年男性IFN-β基因表达量和中老年女性比较,差异无统计学意义(P<0.05)。结论性别和年龄对I型干扰素表达有一定的影响,为临床研究免疫相关性疾病的发病机制提供理论基础。

Objective To explore the effects of gender and age on the expression of typeⅠinterferon by in-vestigating the expression of typeⅠinterferon mRNA in peripheral blood mononuclear cells (PBMC) of healthy indi-viduals. Methods Peripheral blood samples were collected from 113 healthy individuals, which consisted of 54 mid-dle-and old-aged participants [27 females and 27 males, with the mean age of (63±8) years old] and 59 young partici-pants [30 females and 29 males, with the mean age of (36±6) years old]. Expression levels of interferon-α(IFN-α) and interferon-β(IFN-β) mRNA were detected by real-time quantitative PCR. Results The mRNA expression of gene IFN-α and IFN-β were significantly different between healthy middle- and old-aged males and young males (P<0.05), and the gene expression of the former was 25 and 2.9 times lower than the latter, respectively. The mRNA ex-pression of gene IFN-αand IFN-βshowed significant difference between healthy middle-and old-aged females and young females (P<0.05), and the gene expression of the former was 6.67 and 3.57 times lower than the latter, respec-tively. The mRNA expression of gene IFN-αand IFN-βwere significantly different between healthy young males and young females (P<0.05), and the gene expression of the former was 4 and 1.92 times lower than the latter, respective-ly. The mRNA expression of gene IFN-α was significantly different between healthy males and females of middle-and old-age (P<0.05), and the gene expression of the former was 16.67 times lower than the latter. However, the ex-pression of IFN-β gene showed no statistically significant differences between middle- and old-aged males and fe-males (P<0.05). Conclusion Gender and age discrepancies have a relationship with the expression of typeⅠinter-feron, which may provide a theoretical basis for the pathogenesis of clinical studies of immune related diseases.
