目的:探讨海马齿状回(DG)的GABAB 受体是否参与血管性痴呆(VD)大鼠空间学习记忆损害。方法 SD雄性大鼠24只,随机分为假手术组、VD组及GABAB受体阻断剂沙克洛芬组,双侧颈总动脉永久性结扎法制备VD大鼠模型。利用Morris水迷宫检测大鼠的空间学习记忆能力;脑部微量透析法和高效液相色谱法测定DG区γ-氨基丁酸( GABA)含量;免疫组织化学法观察 DG区 GABAB受体表达;用脑部微量注射法往DG区注射GABAB受体阻断剂沙克洛芬,观察其对VD模型大鼠空间学习记忆的影响。结果 VD大鼠海马 DG区GABA 含量明显增加,而 GABAB受体表达减少(均为P<0.05)。 VD大鼠的空间学习记忆能力显著下降,而训练前向DG区微量注射GABAB受体阻断剂可改善VD大鼠的空间学习记忆损害。结论 VD模型大鼠海马DG内增加的GABA通过GABAB受体参与其空间学习记忆的损害。

Objective To investigate whether the GABAB receptors in the hippocampal dentate gyrus ( DG) are were involved in spatial learning and memory impairments in vascular dementia ( VD) .Methods Totally 24 SD rats were randomly divided into sham-opera-ted,VD and GABAB receptor antagonist groups ,and the VD rat model was established by permanent bilateral carotid occlusion .The extracel-lular concentration of GABA in DG was determined by in vivo microdialysis and HPLC ,and spatial learning and memory was assessed by Mor-ris water maze test.The role of GABAB receptors in spatial learning and memory impairments of VD rats was next investigated by microinjec -tion of its antagonists into the DG .Results The extracellular concentration of GABA in DG was increased and the number of GABAB recep -tor-positive cells in DG was decreased in VD group compared with those of sham-operated group .Spatial learning and memory was impaired in VD group,and microinjection of GABAB receptor antagonist into DG significantly attenuated the spatial learning and memory deficits of VD rats.Conclusions Increased GABA in the hippocampal DG might play a role in spatial learning and memory impairment in VD rats via GABAB receptors.
