目的:观察黑逍遥散对Aβ25~35诱导阿尔茨海默病(AD)模型大鼠学习记忆能力改善作用、脑组织乙酰胆碱酯酶(AchE)、胆碱乙酰转移酶和( ChAT)、单胺氧化酶( MAO)及海马病理改变的影响,探讨黑逍遥散防治AD的作用及其机制。方法110只SD大鼠先随机挑选16只为假手术组,余下作为造模组,经过7d的水迷宫实验,剔除记忆力异常的大鼠。造模组再按随机数字表分为模型组、阳性对照组及黑逍遥散高、中、低剂量组。采用Aβ25~35海马区注射诱发大鼠 AD 模型;高、中、低剂量组分别按4.25、8.50和17.0 g? kg-1? d-1灌胃给药,阳性对照组给予哈伯因(0.02 mg? kg-1? d-1)灌胃,每组14只,给药时间持续28 d。 Morris水迷宫检测各组大鼠学习记忆能力的变化,最后一次水迷宫后,处死大鼠,收集大鼠海马及血液样本。 ELISA法测大鼠脑匀浆AchE、ChAT、MAO活性;HE染色法观察海马病理损伤;电镜观察各组大鼠海马组织超微结构的变化。结果黑逍遥散能改善Aβ25~35所致AD模型大鼠行为学功能,提高学习和记忆能力;降低 AD大鼠脑组织AchE 和 MAO活性、升高ChAT 活性( P<0.05);改善Aβ25~35所致AD模型大鼠AD大鼠海马病理损伤。结论黑逍遥散改善AD大鼠学习记忆力、保护AD 模型大鼠脑组织结构和功能,这可能与其调节胆碱能和单胺类神经元神经递质有关。

Objetc ive To explore the influence of Heixiaoyao powder on learning and memory ability ,cerebral neurotransmitters and the pathological changes of hippocampus tissue in Alzheimer's disease(AD) rats induced by Aβ25~35.Methods The SD rats were randomly divided into sham surgery rats and model groups ,and all rats of model group were given water maze tests for 7 days.These eligible rats were randomly divided into model ,positive control ,Heixiaoyao powder high ,medium and low dose groups ,14 rats in each group .Aβ25~35 fragments was injected into the hippocampus to make the rat ’ s AD mode1.All rats of Heixiaoyao powder group were orally administered Heixiaoyao powder at low,medium and high doses (4.25,85.0 ,17.0 g? kg-1? d-1 ),the positive control group was orally given huperzine A solution 0.02 mg? kg-1? d-1 ,model and sham groups were given equal volume double distilled water daily for 28 d.The learning and memory ability of rats were detected by Morris water maze;brain homogenate stration of AchE ,ChAT,MAO were examined by chemistry chromatometry .After the last administration ,the rats were sacrificed and the serum and hippocampus homogenate stration of AchE ,MAO,ChAT were examined by ELISA.Results Heixiaoyao powder improved the behavioral function of AD rats induced by A β25~35 .Compared with those of model group , Heixiaoyao powder obviously decreased the activities of AchE and MAO and increased the active of ChAT in brain tissue (P<0.05).Conclu-sions Heixiaoyao powder could improve the learning and memory ability of AD rats ,and the effect might be related to regulating the levels of cholinergic transmitter and cerebral monoamine neurotransmitters .
