目的:探讨雄性成年SD大鼠心肌缺血30min再灌注120min( I30minR120min)后心肌组织中缺氧诱导因子-1α( hypoxia-inducible factor 1α,HIF-1α)及核转录因子-κB( Nuclear transcription factor-κB,NF-κB)的表达变化。方法将30只SD大鼠分为正常组(Normal)、假手术组(Sham)和缺血30min再灌注120min组(I30minR120min),I30minR120min组结扎雄性成年SD大鼠冠状动脉左前降支, ELISA和Western blot法检测心肌组织中HIF-1α和NF-κB蛋白表达的改变。结果与假手术组相比,I30minR120min组心肌中HIF-1α和NF-κB 的蛋白水平明显增加( P <0.01)。结论心肌缺血再灌注可使心肌HIF-1α和NF-κB的表达增高。

목적:탐토웅성성년SD대서심기결혈30min재관주120min( I30minR120min)후심기조직중결양유도인자-1α( hypoxia-inducible factor 1α,HIF-1α)급핵전록인자-κB( Nuclear transcription factor-κB,NF-κB)적표체변화。방법장30지SD대서분위정상조(Normal)、가수술조(Sham)화결혈30min재관주120min조(I30minR120min),I30minR120min조결찰웅성성년SD대서관상동맥좌전강지, ELISA화Western blot법검측심기조직중HIF-1α화NF-κB단백표체적개변。결과여가수술조상비,I30minR120min조심기중HIF-1α화NF-κB 적단백수평명현증가( P <0.01)。결론심기결혈재관주가사심기HIF-1α화NF-κB적표체증고。
Objective To study the expression of hypoxia-inducible factor 1α( HIF-1α) and nuclear transcription factor-κB( NF-κB) in myocardial ischemia-reperfusion injury of adult male SD rats.Methods 30 adult male SD rats were randomly divid-ed into 3 groups:normal group, sham group and I30minR120min group.SD rats in I30minR120min group were subjected to 30-min myocardial ischemia by ligation of the left anterior descending coronary artery and 120-min reperfusion.The expressions of HIF-1αand NF-κB were measured in myocardium by Western blot and ELISA.Results Compared with the sham group, the levels of both HIF-1αand NF-κB were increased in the ischemic region of myocardium in ischemia 30 min and reperfusion 120 min.Conclusion The expression of HIF-1αand NF-κB in myocardium was increased in myocardial ischemia reperfusion injury.
