目的 探讨人食管鳞状细胞癌中人类黑色素瘤相关抗原A9 (MAGE-A9)的表达及临床意义.方法 应用免疫组织化学检测53例食管鳞癌和30例正常食管组织中MAGE-A9蛋白表达水平,实时定量反转录聚合酶链反应(RT-qPCR)检测食管鳞癌癌组织和正常食管黏膜组织中MAGE-A9基因表达水平.结果 MAGE-A9在正常食管黏膜中微弱表达,而在食管鳞癌组织中显著高表达;癌组织中MAGE-A9蛋白相对表达量(3.620±2.009)明显高于正常食管黏膜组织(0.300±0.652),差异有统计学意义(P<0.05).MAGE-A9mRNA表达水平(0.722±0.352)显著高于正常食管组织(0.223±0.292),差异有统计学意义(P<0.05).结论 MAGE-A9在食管鳞癌中高表达,且与临床病理因素及预后相关.

Objective To investigate the expression of melanoma associated antigen A9 (MAGE-A9) in esophageal squamous cell carcinoma (ESCC) and normal esophageal mucosa tissues,and explore its correlation with clinicopathological characteristics and prognosis.Methods The expression of MAGE-A9 protein in ESCC tissues (n =53) and normal esophageal mucosa tissues (n =30) was detected by immunohistochemistry.Real-time quantitative reverse transcriptase-polymerase chain reaction (RT-qPCR) was used to examine the expression of MAGE-A9 mRNA in fresh ESCC specimens and normal esophageal mucosa tissues.Results The MAGE-A9 positive staining was predominantly localized in the cytoplasm of cancer cells.The expression of MAGE-A9 protein and mRNA was significantly higher in ESCC than in normal tissues (3.620 ±2.009 vs.0.300 ±0.652,P <0.05).Moreover,the expression level of MAGE-A9 protein in ESCC tissues was significantly correlated to smoking (P < 0.05),tumor sizes (P < 0.05),and lymph node metastasis (P < 0.05).Cox regression analysis revealed that MAGE-A9 expression level (P < 0.05) and lymph node metastasis (P < 0.05) were independent prognostic factors of ESCC.Conclusion MAGE-A9 is up-regulated in ESCC.High expression of MAGE-A9 correlates with clinicopathological characteristics and unfavorable survival in ESCC.
