目的 观察RhoC基因表达变化在槲皮素(quercetin)抑制肝细胞癌侵袭转移中的作用.方法 肝癌SF7721细胞经quercetin处理,检测SF7721侵袭迁移能力、RhoC mRNA和蛋白表达;并用quercetin治疗裸鼠移植瘤,观察肝癌生长、肺转移率、RhoC mRNA和蛋白表达变化.结果 quercetin在40、80、160 μmol/L时细胞侵袭迁移力均显著低于对照组.同时发现在24、48、72 h细胞侵袭迁移力均显著低于对照组.40μmol/L至80μmol/L时RhoC蛋白表达量显著下调(0.91±0.10,0.64±0.01,0.66±0.04,P<0.05).当160 μmol/L作用于SF7721细胞时,在24h至48 hRhoC蛋白表达量显著下调(0.54±0.06,0.19±0.01,0.20±0.01,P<0.05);移植瘤quercetin组RhoC蛋白表达显著低于对照组(5.10 ±0.56比1.91±0.59),quercetin组裸鼠移植瘤大小、重量和肺转移率显著低于对照组[分别为(5.78±3.01) cm3比(1.42±1.01) cm3;(7.02±2.30)g比(2.07 ±0.90)g,5/8比0/8,P<0.01)].结论 quercentin可下调肝癌细胞RhoC蛋白表达,抑制肝癌细胞侵袭迁移力,抑制裸鼠移植人肝癌生长和肺转移.

목적 관찰RhoC기인표체변화재곡피소(quercetin)억제간세포암침습전이중적작용.방법 간암SF7721세포경quercetin처리,검측SF7721침습천이능력、RhoC mRNA화단백표체;병용quercetin치료라서이식류,관찰간암생장、폐전이솔、RhoC mRNA화단백표체변화.결과 quercetin재40、80、160 μmol/L시세포침습천이력균현저저우대조조.동시발현재24、48、72 h세포침습천이력균현저저우대조조.40μmol/L지80μmol/L시RhoC단백표체량현저하조(0.91±0.10,0.64±0.01,0.66±0.04,P<0.05).당160 μmol/L작용우SF7721세포시,재24h지48 hRhoC단백표체량현저하조(0.54±0.06,0.19±0.01,0.20±0.01,P<0.05);이식류quercetin조RhoC단백표체현저저우대조조(5.10 ±0.56비1.91±0.59),quercetin조라서이식류대소、중량화폐전이솔현저저우대조조[분별위(5.78±3.01) cm3비(1.42±1.01) cm3;(7.02±2.30)g비(2.07 ±0.90)g,5/8비0/8,P<0.01)].결론 quercentin가하조간암세포RhoC단백표체,억제간암세포침습천이력,억제라서이식인간암생장화폐전이.
Objective To investigate the role of RhoC in the procedure of quercetin depress the invasion and metastasis of hepatocellular carcinoma.Methods Add quercetin in the medium of SF7721 cell then test the invasion and metastasis of SF7721 cells,and detect RhoC mRNA and protein expression of the cells.After that treat the nude mouse model of human hepatocellular carcinoma in situ with quercetin,measure the growth,pulmonary metastasis rate,RhoC mRNA and protein expression of the tumour.Results The invasion and metastasis of SF7721 cells are significantly lower than that of control,when quercetin concentration are 40,80 and 160 μmol/L.And at the time of 24,48 and 72 h,the invasion and metastasis of SF7721 cells are prominently weaker than that of control.RhoC protein expression is significantly depressed (semiquantitative is 0.91 ± 0.10 vs.0.64 ± 0.01,0.66 ± 0.04),when quercetin concentration is from 40 μmol/L to 80 μnol/L (P < 0.05).And the same to the time of 24 h to 48 h (semiquantitative is 0.54 ±0.06 vs.0.19 ±0.01,0.20 ±0.01,P<0.05).RhoC protein expression of transplanted tumour from quercetin group is remarkable lower than that of control (semiquantitative is 5.10 ± 0.56 vs.1.91 ± 0.59).The size,weight and pulmonary metastasis rate of the tumour from quercetin group are marked lower than that of control[respectively is (5.78 ±3.01) cm3 vs.(1.42 ± 1.01) cm3;(7.02 ± 2.30) g vs.(2.07 ± 0.90) g,5/8 vs.0/8,P < 0.05].Conclusion Quercetin can significantly down-regulate RhoC protein expression of hepatocellular carcinoma cells,depress invasion and migration of the cells,and inhibit the growth and pulmonary metastasis of human hepatocellular carcinoma transplanted into mouse liver.
