目的:探讨P物质对星形胶质细胞的直接作用。方法利用原代培养的星形胶质细胞,P物质孵育后,采用免疫细胞化学的方法观察细胞形态的变化,采用细胞内钙离子成像方法记录P物质作用前后胞内钙荧光强度的变化。采用酶联免疫吸附试验检测细胞释放白细胞介素(IL)-10的情况。结果 P物质孵育后,星形胶质细胞表现出激活态的形态学改变,P物质孵育后引起星形胶质细胞内Ca2+浓度增加,星形胶质细胞释放抗炎因子IL-10减少。结论脊髓水平传递痛信号的神经递质P物质可刺激活化脊髓星形胶质细胞,活化的星形胶质细胞通过抑制抗炎因子IL-10的释放,最终导致慢性疼痛的发生。

목적:탐토P물질대성형효질세포적직접작용。방법이용원대배양적성형효질세포,P물질부육후,채용면역세포화학적방법관찰세포형태적변화,채용세포내개리자성상방법기록P물질작용전후포내개형광강도적변화。채용매련면역흡부시험검측세포석방백세포개소(IL)-10적정황。결과 P물질부육후,성형효질세포표현출격활태적형태학개변,P물질부육후인기성형효질세포내Ca2+농도증가,성형효질세포석방항염인자IL-10감소。결론척수수평전체통신호적신경체질P물질가자격활화척수성형효질세포,활화적성형효질세포통과억제항염인자IL-10적석방,최종도치만성동통적발생。
Objective To investigate the effect of substance P on the spinal astrocytes. Methods Primary cul-tured astrocytes was exposed to substance P. Changes in the morphology of astrocytes were assessed by immunocyto-chemistry assay, intracellular calcium concentration was observed by calcium imaging. The release of interleukin-10 (IL-10) was measured with enzyme-linked immunosorbent assay. Results Substance P induced the morphological change of astrocytes. Substance P incubation increased intracellular calcium concentration in astrocytes, and inhibited the release of IL-10. Conclusion Spinal neurotransmitter substance P can activate astrocytes. Activated astrocytes in-hibited the anti-inflammatory cytokine IL-10 release, and were involved in the pathology of chronic pain.
