目的:研究柔红霉素(Daunorubicin,DNR)和枸橼酸他莫昔芬(Tamoxifen Citrate,TMX)对人乳腺癌细胞MCF-7的协同作用。方法:取对数生长期MCF-7细胞,分为柔红霉素给药组(浓度范围0.25~1.5μmol/L)、枸橼酸他莫昔芬给药组(浓度范围2.5~15μmol/L)和两药混合给药组,采用MTT法考察各给药组作用24h后对MCF-7细胞的增长抑制作用,利用中效原理计算合用指数来判定DNR和TMX的协同作用。结果:联合给药组的细胞抑制率均高于单独给药组,并且IC50时联合给药组的柔红霉素和枸橼酸他莫昔芬剂量低于单独给药组剂量。两药合用时(DNR∶TMX=1∶10,摩尔比),当DNR剂量≥0.188mmol/L,TMX≥1.887mmol/L时,合用指数CI<1,二者表现为协同作用。结论:柔红霉素和枸橼酸他莫昔芬联合应用能抑制人乳腺癌细胞MCF-7生长,在一定条件下,两者为协同作用,并且随着药物剂量增加,效应增强,协同作用呈现更强。

목적:연구유홍매소(Daunorubicin,DNR)화구연산타막석분(Tamoxifen Citrate,TMX)대인유선암세포MCF-7적협동작용。방법:취대수생장기MCF-7세포,분위유홍매소급약조(농도범위0.25~1.5μmol/L)、구연산타막석분급약조(농도범위2.5~15μmol/L)화량약혼합급약조,채용MTT법고찰각급약조작용24h후대MCF-7세포적증장억제작용,이용중효원리계산합용지수래판정DNR화TMX적협동작용。결과:연합급약조적세포억제솔균고우단독급약조,병차IC50시연합급약조적유홍매소화구연산타막석분제량저우단독급약조제량。량약합용시(DNR∶TMX=1∶10,마이비),당DNR제량≥0.188mmol/L,TMX≥1.887mmol/L시,합용지수CI<1,이자표현위협동작용。결론:유홍매소화구연산타막석분연합응용능억제인유선암세포MCF-7생장,재일정조건하,량자위협동작용,병차수착약물제량증가,효응증강,협동작용정현경강。
Objective:To study the synergistic effect of daunorubicin combined with tamoxifen on breast cancer cells MCF-7 with the Chou-Talalay method. Methods: The effect of daunorubicin or tamoxifen alone and their combination on MCF-7 cell proliferation were measured by MTT assay. The median-effect principle was used to evaluate the synergistic effect.Results: The combination of daunorubicin and tamoxifen had synergistic effect on MCF-7 cell, when DNR≥0.188mmol/L and TMX≥1.887mmol/L. Conclusion:Daunorubicin combined with tamoxifen has inhibitory effect on MCF-7 cell proliferation, and the interactive effect of the two drugs in combination is synergistic under certain conditions.
