以马铃薯叶片为试验材料,采用改良CTAB法和试剂盒法提取马铃薯叶片基因组DNA。结果表明,2种方法均可得到满足SSR-PCR扩增要求的DNA,其中试剂盒较改良CTAB法提取的DNA质量高、杂质少,DNA得率低,操作简单,耗时短,毒害小,但费用较改良CTAB法高。实践中可根据条件选择适宜方法提取马铃薯基因组DNA。

The potato leaves as test materials,improved CTAB method and plant genomic DNA extraction Kit is used to extract genomic DNA from leaves of potato in field respectively. The result shows that DNA extacted by the two methods also met demands for SSR-PCR amplification;The quality of DNA obtained by plant genomic DNA Kit is better but the achievement rate of DNA is lower. Moreover,the method of plant genomic DNA Kit is much easier,needed shorter time,had lower toxicity,and high cost than improved CATB method. Suitable method can be choose to genomic DNA of potato according to the actual conditions of the laboratory.
