目的:探讨温阳化浊通络方含药血清对Wnt-1诱导的人皮肤成纤维细胞表型转化及迁移的影响。方法:通过对Wistar大鼠灌胃制备温阳化浊通络方含药血清,体外培养正常人皮肤成纤维细胞,分为空白对照组、Wnt组和温阳化浊通络方组,作用24,48,72 h后,分别用RT-PCR和Western blot法检测α-SMA mRNA及蛋白的表达,Transwell试验检测成纤维细胞迁移情况。结果:与空白对照组比较, Wnt组在不同作用时间下能诱导人皮肤成纤维细胞α-SMA mRNA及蛋白表达(P<0.01),增加其迁移数量(P<0.01);与Wnt组比较,温阳化浊通络方组在不同作用时间下能够显著抑制Wnt-1诱导的人皮肤成纤维细胞α-SMA mRNA及蛋白表达(P<0.05,P<0.01),同时阻止其迁移(P<0.01)。结论:温阳化浊通络方含药血清能抑制Wnt-1诱导的人皮肤成纤维细胞表型转化及迁移,具有阻止或延缓系统性硬化病纤维化的潜能。

목적:탐토온양화탁통락방함약혈청대Wnt-1유도적인피부성섬유세포표형전화급천이적영향。방법:통과대Wistar대서관위제비온양화탁통락방함약혈청,체외배양정상인피부성섬유세포,분위공백대조조、Wnt조화온양화탁통락방조,작용24,48,72 h후,분별용RT-PCR화Western blot법검측α-SMA mRNA급단백적표체,Transwell시험검측성섬유세포천이정황。결과:여공백대조조비교, Wnt조재불동작용시간하능유도인피부성섬유세포α-SMA mRNA급단백표체(P<0.01),증가기천이수량(P<0.01);여Wnt조비교,온양화탁통락방조재불동작용시간하능구현저억제Wnt-1유도적인피부성섬유세포α-SMA mRNA급단백표체(P<0.05,P<0.01),동시조지기천이(P<0.01)。결론:온양화탁통락방함약혈청능억제Wnt-1유도적인피부성섬유세포표형전화급천이,구유조지혹연완계통성경화병섬유화적잠능。
Objective:To investigate the effects of Wenyang Huazhuo Tongluo Prescription(温阳化浊通络方)on the phenotype transformation and migration of human skin ifbroblast induced by Wnt-1.Methods:Medicated serum of Wistar rats was got by intragastric administration of Wenyang Huazhuo Tongluo Prescription and normal human skin fibroblasts were cultured in vitro.The rats were divided into a blank control group,a Wnt group and a Wenyang Huazhuo Tongluo Prescription group.Respectively after 24,48,72 hours,RT-PCR and Western blot were used to detect the expressions of α-SMA mRNA and protein,and Transwell was used to detect the migration of fibroblasts.Results:Compared with the blank control group,human skin fibroblasts a-SMA mRNA and protein expression in the Wnt group could be induced in different time(P<0.01),and the number of migration increased(P<0.01).Compared with the Wnt group,human skin ifbroblasts a-SMA mRNA and protein expression in the Wenyang Huazhuo Tongluo Prescription group were obviously inhibited(P < 0.05, P<0.01) and their migration were prevented(P<0.01).Conclusion:Medicated serum of Wenyang Huazhuo Tongluo Prescription can inhibit the phenotype transformation and migration of human skin ifbroblast induced by Wnt-1,having the potential to prevent or delay systemic sclerosis ifbrosis.
