目的:探讨细胞毒 T 淋巴细胞抗原4(cytotoxic T lymphocyte antigen 4,CTLA-4)-318 T/C、CT60 G/A、+49 G/A 基因多态性与宫颈癌之间的相关性。方法选取2012年12月至2014年7月292例宫颈癌患者(患者组),及同期355例健康体检者(对照组),采集研究对象外周静脉血5 ml,抽提全血基因组 DNA,PCR 扩增后直接测序法检测 CTLA-4-318 T/C,CT60 G/A,+49 G/A 基因多态性的分布。结果Logistic 回归分析显示,-318CC 和 CT60 AA 基因型均与宫颈癌发病相关(P <0.05)。与对照组比较,携带 CTLA-4-318T、CT60 g 等位基因的宫颈癌患者明显增加(均 P <0.05);携带 CTLA-4+49 gG 基因型、+49A 等位基因的宫颈癌患者,差异无统计学意义(P >0.05)。结论CTLA-4-318、CT60基因多态性与宫颈癌发病有关。

목적:탐토세포독 T 림파세포항원4(cytotoxic T lymphocyte antigen 4,CTLA-4)-318 T/C、CT60 G/A、+49 G/A 기인다태성여궁경암지간적상관성。방법선취2012년12월지2014년7월292례궁경암환자(환자조),급동기355례건강체검자(대조조),채집연구대상외주정맥혈5 ml,추제전혈기인조 DNA,PCR 확증후직접측서법검측 CTLA-4-318 T/C,CT60 G/A,+49 G/A 기인다태성적분포。결과Logistic 회귀분석현시,-318CC 화 CT60 AA 기인형균여궁경암발병상관(P <0.05)。여대조조비교,휴대 CTLA-4-318T、CT60 g 등위기인적궁경암환자명현증가(균 P <0.05);휴대 CTLA-4+49 gG 기인형、+49A 등위기인적궁경암환자,차이무통계학의의(P >0.05)。결론CTLA-4-318、CT60기인다태성여궁경암발병유관。
Objective To explore the correlation between cytotoxic T lymphocyte antigen 4(CTLA-4) genotypes-318 T/C, CT60 G/A,+49 G/A and occurrence of cervical cancer .Methods Two hundred and ninety-two patients with cervical cancer from De -cember 2012 to July 2014 were selected as study group,and 355 cases of healthy controls were recruited as control group .Peripheral blood of each participant was collected and genomic DNA was extracted .The distribution of CTLA-4-318 T/C,CT60 G/A,+49 G/A genotypes were detected by direct sequencing .Results The CTLA-4-318 CC and CT60 AA genotypes were correlated with the decrease of cervical cancer risk(P <0.05).Compared with control group,the frequencies of CTLA-4-318T allele and CT60 g allele carriers were significantly higher in the cancer patients (P <0.05).No significant differences were found in the genotype and allele frequencies in +49 G/A between patients and healthy controls (P >0.05).Conclusion The polymorphisms of CTLA-4-318 and CT60 are correlated with cervical cancer risk.
