目的:探讨通心络对糖尿病足大鼠内皮细胞功能指标[一氧化氮(NO)、内皮素(ET)、血管性假血友病因子(vWF)],内皮细胞的形态及ET、内皮型一氧化氮合酶(eNOS)、诱生型一氧化氮合酶(iNOS)表达的影响及其可能机制。方法健康SD大鼠45只,分为对照组、模型组及通络组,各15只。采用高脂喂养、链脲佐菌素( STZ)和结扎并剪断SD大鼠股动脉和股静脉的方法复制糖尿病足大鼠模型,对照组、模型组给予以生理盐水灌胃,通络组给予通心络治疗,共6周。生化法检测血清NO、ET、vWF水平,光镜、透射电镜观察血管内皮细胞的形态和超微结构变化,免疫组化观察ET1、eNOS、iNOS的表达。结果与对照组比较,模型组NO水平降低, ET、vWF 水平升高( P <0.05, P <0.01);与模型组比较,通络组糖尿病足大鼠血清NO增加、血浆ET、vWF降低( P <0.05, P <0.01),内皮细胞形态有明显改善,eNOS 表达增加,ET、iNOS表达减少。结论通心络治疗能改善糖尿病足大鼠血管内皮细胞的损伤和功能障碍。

목적:탐토통심락대당뇨병족대서내피세포공능지표[일양화담(NO)、내피소(ET)、혈관성가혈우병인자(vWF)],내피세포적형태급ET、내피형일양화담합매(eNOS)、유생형일양화담합매(iNOS)표체적영향급기가능궤제。방법건강SD대서45지,분위대조조、모형조급통락조,각15지。채용고지위양、련뇨좌균소( STZ)화결찰병전단SD대서고동맥화고정맥적방법복제당뇨병족대서모형,대조조、모형조급여이생리염수관위,통락조급여통심락치료,공6주。생화법검측혈청NO、ET、vWF수평,광경、투사전경관찰혈관내피세포적형태화초미결구변화,면역조화관찰ET1、eNOS、iNOS적표체。결과여대조조비교,모형조NO수평강저, ET、vWF 수평승고( P <0.05, P <0.01);여모형조비교,통락조당뇨병족대서혈청NO증가、혈장ET、vWF강저( P <0.05, P <0.01),내피세포형태유명현개선,eNOS 표체증가,ET、iNOS표체감소。결론통심락치료능개선당뇨병족대서혈관내피세포적손상화공능장애。
Objective To investigate the effect of Tongxinluo capsule on diabetic foot rat endothelial function index:nitric oxide ( NO) , endothelin ( ET) , vascular von Willebrand factor ( vWF) and endothelial cell morphology and ET ,endo-thelial nitric oxide synthase (eNOS) and inducible nitric oxide synthase (iNOS), and its possible mechanism.Methods Forty-five healthy SD rats were divided into control group , model group with 15 rats in each group.high-fat feeding, streptozo-tocin ( STZ) , ligation and cut the femoral artery and femoral vein of SD rat to establish diabetic foot rat model , control group , model group were given normal saline gavage , Tongluo group was given Tongxinluo capsule treatment for 6 weeks.Serum lev-els of NO, ET, and vWF were detected by biochemical method .The morphology and ultrastructure of vascular endothelial cells were observed by light microscope and transmission electron microscope .The expression of eNOS , iNOS and ET were ob-served by immunohistochemistry .Results Compared to the control group , model group ’ s NO level was decreased , ET and vWF levels were elevated ( P <0.05, P <0.01);compared with the model group , Tongluo group diabetic foot rats ’ level of NO was increased, plasma ET and vWF were decreased ( P <0.05, P <0.01), the morphology of endothelial cells were sig-nificantly improved , and eNOS expression was increased , ET and iNOS expression was reduced .Conclusion Tongxinluo capsule can improve diabetic foot rat vascular endothelial cell from injury and dysfunction .
