目的:探讨乌鸡黑色素对1-甲基-4-苯基-1,2,3,6-四氢吡啶( MPTP)致帕金森病( PD)模型小鼠的神经保护作用及其机制。方法2013年3月—2014年5月,按照随机数字表法将60只雄性C57/B6小鼠分为对照组(n=10)、 MPTP模型组(n=12)和乌鸡黑色素治疗组(n=38);乌鸡黑色素治疗组又进一步分为10 mg/kg治疗组(n=13)、30 mg/kg治疗组(n=13)和100 mg/kg治疗组(n=12)。对照组小鼠采用0.9%氯化钠溶液灌胃。MPTP模型组和乌鸡黑色素治疗组小鼠腹腔注射MPTP (30 mg/kg,1次/d,共7 d),同时乌鸡黑色素治疗组小鼠分别灌胃给予不同剂量的乌鸡黑色素(10 mg/kg、30 mg/kg和100 mg/kg,1次/d,共7 d)。剔除建模不成功小鼠,最后实际入组小鼠分别为对照组10只、 MPTP模型组10只、10 mg/kg治疗组10只、30 mg/kg治疗组9只、100 mg/kg治疗组9只。比较各组小鼠黑质酪氨酸羟化酶( TH)免疫阳性纤维的表达变化、黑质致密部TH免疫阳性神经元数及纹状体TH水平。结果对照组可见大量呈束状、密集、排列规则有序TH免疫阳性神经纤维; MPTP模型组TH免疫阳性神经纤维稀疏、断裂且排列零散无序;乌鸡黑色素治疗组与模型组染色结果相比,仍可见呈束状、较密集、排列较有序的TH免疫阳性神经纤维,尤其以30 mg/kg治疗组最为明显。 MPTP模型组、10 mg/kg治疗组、30 mg/kg治疗组、100 mg/kg治疗组黑质致密部TH免疫阳性神经元数低于对照组( P<0.05);30 mg/kg治疗组黑质致密部TH免疫阳性神经元数高于MPTP模型组(P<0.05)。 MPTP模型组、10 mg/kg治疗组、30 mg/kg治疗组、100 mg/kg治疗组纹状体TH水平低于对照组(P<0.05);30 mg/kg治疗组、100 mg/kg治疗组纹状体TH水平高于MPTP模型组(P<0.05)。结论乌鸡黑色素灌胃给药能减缓MPTP诱导的亚急性PD模型小鼠黑质多巴胺能神经元的死亡和纹状体TH免疫阳性神经纤维的丢失,提示口服乌鸡黑色素对于PD模型小鼠有潜在的神经保护作用。

목적:탐토오계흑색소대1-갑기-4-분기-1,2,3,6-사경필정( MPTP)치파금삼병( PD)모형소서적신경보호작용급기궤제。방법2013년3월—2014년5월,안조수궤수자표법장60지웅성C57/B6소서분위대조조(n=10)、 MPTP모형조(n=12)화오계흑색소치료조(n=38);오계흑색소치료조우진일보분위10 mg/kg치료조(n=13)、30 mg/kg치료조(n=13)화100 mg/kg치료조(n=12)。대조조소서채용0.9%록화납용액관위。MPTP모형조화오계흑색소치료조소서복강주사MPTP (30 mg/kg,1차/d,공7 d),동시오계흑색소치료조소서분별관위급여불동제량적오계흑색소(10 mg/kg、30 mg/kg화100 mg/kg,1차/d,공7 d)。척제건모불성공소서,최후실제입조소서분별위대조조10지、 MPTP모형조10지、10 mg/kg치료조10지、30 mg/kg치료조9지、100 mg/kg치료조9지。비교각조소서흑질락안산간화매( TH)면역양성섬유적표체변화、흑질치밀부TH면역양성신경원수급문상체TH수평。결과대조조가견대량정속상、밀집、배렬규칙유서TH면역양성신경섬유; MPTP모형조TH면역양성신경섬유희소、단렬차배렬령산무서;오계흑색소치료조여모형조염색결과상비,잉가견정속상、교밀집、배렬교유서적TH면역양성신경섬유,우기이30 mg/kg치료조최위명현。 MPTP모형조、10 mg/kg치료조、30 mg/kg치료조、100 mg/kg치료조흑질치밀부TH면역양성신경원수저우대조조( P<0.05);30 mg/kg치료조흑질치밀부TH면역양성신경원수고우MPTP모형조(P<0.05)。 MPTP모형조、10 mg/kg치료조、30 mg/kg치료조、100 mg/kg치료조문상체TH수평저우대조조(P<0.05);30 mg/kg치료조、100 mg/kg치료조문상체TH수평고우MPTP모형조(P<0.05)。결론오계흑색소관위급약능감완MPTP유도적아급성PD모형소서흑질다파알능신경원적사망화문상체TH면역양성신경섬유적주실,제시구복오계흑색소대우PD모형소서유잠재적신경보호작용。
Objective To investigate the neuroprotective effects of melanine derived from black -bone silky fowl ( BSF) on model mouse with MPTP-induced Parkinson′s disease ( PD ) and the mechanism.Methods From March 2013 to May 2014, 60 C57/B6 rats were divided into control group (n=10), MPTP group (n=12) and melanine group (n=38) by using random number table; the melanine group was further divided into 10 mg/kg group ( n=13) , 30 mg/kg group ( n=13) and 100 mg/kg group (n=12) .Rats in the control group were administrated with 0.9% sodium chloride solution by gavage;rats in the MPTP group and melanine group were treated with intraperitoneal injection of MPTP (30 mg/kg; one time per day for 7 days); rats in the melanine group were given BSF melanine of different doses ( 10 mg/kg, 30 mg/kg and 100 mg/kg; one time per day for 7 days) by gavage.Rats with unsuccessful model building were excluded, and at last 10 rats in the control group, 10 in the MPTP group, 10 in the 10 mg/kg group, 9 in the 30 mg/kg group and 9 in the 100 mg/kg group were included in the study.Comparison was made among these groups in the expression changes of tyrosine hydroxylase ( TH) -immunoreactive fiber in substantia nigra ( SN) , the number of TH-immunoreactive neurons in the compact part of SN and the protein level of TH corpus striatum.Results In the control group, plenty of TH-immunoreactive fiber was in fasciculation and in a dense and orderly form; in the MPTP model group, TH-immunoreactive nerve fiber was in a sparse, broken and disordered form; in the melanine group, TH -immunoreactive nerve fiber was in fasciculation and in a dense and orderly form, especially in the 30 mg/kg group.The MPTP group, 10 mg/kg group, 30 mg/kg group and 100 mg/kg group were lower ( P<0.05 ) than the control group in the number of TH-immunoreactive neurons in the compact part of SN; the 30 mg/kg group was higher ( P<0.05) than the MPTP group in the number of TH-immunoreactive neurons in the compact part of SN.The MPTP group, 10 mg/kg group, 30 mg/kg group and 100 mg/kg group were lower ( P<0.05) than the control group in the protein level of TH corpus striatum; the 30 mg/kg group and 100 mg/kg group were higher ( P<0.05 ) than the MPTP group in the protein level of TH corpus striatum.Conclusion The intragastric administration of BSF melanine can reduce the death of substantia nigra DA neurons and the loss of corpus striatum TH -immunoreactive nerve fiber in model rats with MPTP -induced subacute PD.The oral administration of BFS melanine has potential neuroprotective effects on PD model rats.
