目的:探讨复方通莲汤对胃癌MGC803细胞荷瘤裸鼠的抑瘤作用及对缺氧诱导因子-1α(HIF-1α)表达的影响,为该方临床应用提供实验依据。方法胃癌细胞株MGC803接种于裸鼠右前肢皮下。模型组灌胃生理盐水溶液10mL/kg,阳性对照组灌胃消癌平片1.8g(生药量)/kg的药液10mL/kg,复方通莲汤高、中、低剂量组分别灌胃1.5,0.75,0.375 g (生药量)/L的药液10 mL/kg ,各组连续给药3周,再连续观察3周,6周后处死小鼠,分离瘤组织,计算抑瘤率,免疫组织化学法检测肿瘤组织HIF-1α表达。结果高剂量通莲汤显著抑瘤生长,瘤重(1.51±0.49) g ,抑瘤率为(61.04±4.79)%,与模型组比较差异有统计学意义( P<0.05);通莲汤可显著抑制HIF-1α表达,与模型组比较,阳性表达率χ2=4.286,P=0.038<0.05。结论通莲汤可抑制裸鼠人胃癌MGC803细胞移植瘤生长,其抑瘤机制与下调肿瘤细胞HIF-1α蛋白表达有关。

목적:탐토복방통련탕대위암MGC803세포하류라서적억류작용급대결양유도인자-1α(HIF-1α)표체적영향,위해방림상응용제공실험의거。방법위암세포주MGC803접충우라서우전지피하。모형조관위생리염수용액10mL/kg,양성대조조관위소암평편1.8g(생약량)/kg적약액10mL/kg,복방통련탕고、중、저제량조분별관위1.5,0.75,0.375 g (생약량)/L적약액10 mL/kg ,각조련속급약3주,재련속관찰3주,6주후처사소서,분리류조직,계산억류솔,면역조직화학법검측종류조직HIF-1α표체。결과고제량통련탕현저억류생장,류중(1.51±0.49) g ,억류솔위(61.04±4.79)%,여모형조비교차이유통계학의의( P<0.05);통련탕가현저억제HIF-1α표체,여모형조비교,양성표체솔χ2=4.286,P=0.038<0.05。결론통련탕가억제라서인위암MGC803세포이식류생장,기억류궤제여하조종류세포HIF-1α단백표체유관。
Objective The purpose of the research is to observe inhibition of Tonglian Decoction (TD ) on nude mice mod-eled with gastric cancer cells MGC-803 and on hypoxia inducible factor-1α(HIF-1α) expression in tumor tissue ,to provide experimental basis for clinical application of TD .Methods MGC-803 cells are inoculated subcutaneously in right forelimbs of all mice .Mice in model group are treated with saline solution 10 mL/kg orally .Mice in positive control group are treated with Xiaoaiping tablet ,dosage of 10 mL/kg ,1 .8 g/kg (crude drug ) of liquid .Mice in TD groups are subdivided as high (1 . 5 g/L ) ,medium (0 .75 g/L ) and low (0 .375 g/L ) dose group of drug liquid ,with dosage of 10 mL/kg .All mice take ad-ministration for 3 weeks and are observed for another 3 weeks continuously .Tumor volumes are measured for 6 weeks every-day from the beginning of MGC803 cells transplanted .All mice are killed for isolation of tumor tissue ,inhibition ratio of tu-mor-weight measured .Finally ,HIF-1α expression is examined with immunohistochemistry .Results Tumor weight in TD high-dose group is (1 .51 ± 0 .49 ) g ,and the inhibition ratio is (61 .04 ± 4 .79 )% ,which is significantly different from that in the model group ( P<0 .05 );Positive rate of HIF-1αexpression also decreased significantly ,showing statistical difference from the model group (χ2=4 .286 ,P=0 .038<0 .05 ) .Conclusion TD inhibits tumor growth on nude mice modeled with MGC803 ,through downregulation of HIF-1αprotein expression .
