目的:研究川芎嗪对肺癌PG-BE1干细胞样细胞ABCG-2蛋白表达的影响。方法采用无血清成球培养法分选PG-BE1中的干细胞样细胞亚群,并检验其干性,采用MTT法检测PG-BE1干细胞样细胞与原代细胞对常用化疗药的耐药性。采用Western blot法检测PG-BE1干细胞样细胞与原代细胞ABCG-2蛋白表达及不同浓度川芎嗪对ABCG-2蛋白表达的影响。结果 PG-B1干细胞样细胞的干细胞相关蛋白表达及克隆灶形成数目与原代细胞差异有统计学意义;PG-BE1干细胞样细胞化疗药耐药性强于原代细胞;PG-BE1干细胞样细胞ABCG蛋白表达高于原代细胞;川芎嗪对ABCG蛋白的表达有抑制作用。结论无血清成球培养法可有效富集PG-BE1干细胞样细胞,PG-BE1干细胞较其原代细胞有更强的耐药性,这可能与其高表达ABCG-2蛋白有关,川芎嗪对PG-BE1干细胞样细胞ABCG-2蛋白有抑制作用。

목적:연구천궁진대폐암PG-BE1간세포양세포ABCG-2단백표체적영향。방법채용무혈청성구배양법분선PG-BE1중적간세포양세포아군,병검험기간성,채용MTT법검측PG-BE1간세포양세포여원대세포대상용화료약적내약성。채용Western blot법검측PG-BE1간세포양세포여원대세포ABCG-2단백표체급불동농도천궁진대ABCG-2단백표체적영향。결과 PG-B1간세포양세포적간세포상관단백표체급극륭조형성수목여원대세포차이유통계학의의;PG-BE1간세포양세포화료약내약성강우원대세포;PG-BE1간세포양세포ABCG단백표체고우원대세포;천궁진대ABCG단백적표체유억제작용。결론무혈청성구배양법가유효부집PG-BE1간세포양세포,PG-BE1간세포교기원대세포유경강적내약성,저가능여기고표체ABCG-2단백유관,천궁진대PG-BE1간세포양세포ABCG-2단백유억제작용。
Objective To study on the effect of TMP ,which is the effective composition of Chuanxiong ,on the expression of ABCG-2 protein of high metastatic human lung cancer(PG-BE1) cells .Methods First of all ,we sort the cancer stem cell (CSC ) subgroup from the high metastatic human lung cancer cells (PG-BE1 ) by serum free sphere culture method and ex-amine the properties of stem cells .MTT is used to exam the resistance to common chemotherapy medicine of the PG-BE1 CSC like cells and parental cells .Then we use the western blot experiment to verify the expression of ABCG-2 protein in the PG-BE1 CSC like cells and parental cells and the influence of different concentration of TMP on the expression of ABCG-2 protein .Results The number of clone formation and the expression of CSC related protein show significant differences .PG-BE1 CSC cells have stronger chemotherapy resistance than parental cells .The expression of ABCG-2 protein is higher than parental cells .TMP can inhibit the expression of ABCG-2 protein .Conclusion The serum free sphere culture method can effective enrich the CSC like cells and the PG-BE1 CSC like cells show stronger chemothrepy resistance than their parental cells .High expression of ABCG-2 protein may contribute to the themotherapy resistance .TMP can inhibit the expression of the ABCG-2 protein of PG-BE1 CSC like cells .
