目的:探讨金龙蛇颗粒对缺氧状态下胃癌BGC-823细胞上皮间质转化(EMT )的影响并初步研究该过程的相关机理。方法将胃癌BGC-823细胞分成3组:常氧对照组、缺氧对照组(加入150μmol/L的CoCl2)、金龙蛇颗粒含药血清组。观察3组细胞形态变化;RT-PCR检测3组细胞E-cad、N-cad、Vimentin mRNA表达变化。结果常氧对照组的胃癌BGC-823细胞排列较致密,为上皮样表现;缺氧对照组大部分细胞排列较前松散,形态较前拉长,呈间质样表现;金龙蛇颗粒含药血清组介于二者之间。金龙蛇颗粒含药血清组与缺氧对照组相比,E-cad mRNA表达升高,N-cad、Vimentin mRNA表达降低,差异有显著统计学意义( P<0.01)。结论金龙蛇颗粒可抑制缺氧环境中胃癌BGC-823细胞的上皮间质转化,可能与其下调N-cad、Vimentin mRNA表达,升高E-cad mRNA表达相关。

목적:탐토금룡사과립대결양상태하위암BGC-823세포상피간질전화(EMT )적영향병초보연구해과정적상관궤리。방법장위암BGC-823세포분성3조:상양대조조、결양대조조(가입150μmol/L적CoCl2)、금룡사과립함약혈청조。관찰3조세포형태변화;RT-PCR검측3조세포E-cad、N-cad、Vimentin mRNA표체변화。결과상양대조조적위암BGC-823세포배렬교치밀,위상피양표현;결양대조조대부분세포배렬교전송산,형태교전랍장,정간질양표현;금룡사과립함약혈청조개우이자지간。금룡사과립함약혈청조여결양대조조상비,E-cad mRNA표체승고,N-cad、Vimentin mRNA표체강저,차이유현저통계학의의( P<0.01)。결론금룡사과립가억제결양배경중위암BGC-823세포적상피간질전화,가능여기하조N-cad、Vimentin mRNA표체,승고E-cad mRNA표체상관。
Objective To investigate the effects of Jinlongshe granules on EMT of gastric cancer cell BGC-823 under the mi-croenvironment of hypoxia ,and to explore the mechanism of the process .Methods The cells were divided into three groups:normal control group (cultured under normal condition );hypoxia control group (cultured with CoCl2 );Jinlongshe granules group (co-cultured with CoCl2 and Chinese herbal medicine serum ) .Microscopy was used to observe the different morphological change among the three groups and E-cad ,N-cad ,Vimentin mRNA were detected by RT-PCR .Results The cells of normal control group became close together like epithelial features;the cells of hypoxia control group became loose like mesenchymal features;the result of Jinlongshe granules group was between the other two groups .RT-PCR showed that the E-cad mRNA of Jinlongshe granules group was raised significant and the N-cad ,Vimentin mRNA of Jinlongshe granules group was reduced significant compared with hypoxia control group ( P<0 .01 ) .Conclusion Jinlongshe granules can in-hibit the EMT of gastric cancer cell BGC-823 under the microenvironment of hypoxia .The mechanism maybe related to the reduction of N-cad ,Vimentin mRNA and raise of E-cad mRNA .
