目的::观察链脲佐菌素(STZ)诱导的 BALB/c 糖尿病小鼠睾丸微循环变化。方法:20只 BALB/c 小鼠,采用完全随机法分为糖尿病组和对照组,每组10只。糖尿病组小鼠连续5天腹腔注射40mg/kg STZ 诱导糖尿病模型,对照组注射柠檬酸缓冲液。一周后,应用激光多普勒成像系统(Moor LDLS)检测两组小鼠下腹-外阴部皮肤微循环血流灌注水平;分离腹膜,暴露睾丸,应用激光多普勒血流灌注检测系统(Moor VMS-LDF)检测两组小鼠睾丸微循环血流量及睾丸微血管自律运动。心脏灌流后取两组小鼠睾丸制作组织切片,HE 染色观察睾丸微血管形态,免疫组织化学染色两步法观察睾丸微血管内皮细胞抗血小板内皮细胞黏附分子-1(PECAM-1)表达水平。结果:糖尿病组小鼠下腹-外阴部皮肤总血流灌注量低于对照组(P <0.01);睾丸平均血流灌注水平和微血管自律运动频率和振幅均显著低于对照组(P <0.01);糖尿病组小鼠生精上皮受损,成熟生精细胞减少;睾丸间质微血管增多。睾丸间质微血管内皮细胞 PECAM-1表达水平低于对照组(P <0.01)。结论:STZ 诱导的 BALB/c 糖尿病小鼠睾丸微循环存在较多损害。

목적::관찰련뇨좌균소(STZ)유도적 BALB/c 당뇨병소서고환미순배변화。방법:20지 BALB/c 소서,채용완전수궤법분위당뇨병조화대조조,매조10지。당뇨병조소서련속5천복강주사40mg/kg STZ 유도당뇨병모형,대조조주사저몽산완충액。일주후,응용격광다보륵성상계통(Moor LDLS)검측량조소서하복-외음부피부미순배혈류관주수평;분리복막,폭로고환,응용격광다보륵혈류관주검측계통(Moor VMS-LDF)검측량조소서고환미순배혈류량급고환미혈관자률운동。심장관류후취량조소서고환제작조직절편,HE 염색관찰고환미혈관형태,면역조직화학염색량보법관찰고환미혈관내피세포항혈소판내피세포점부분자-1(PECAM-1)표체수평。결과:당뇨병조소서하복-외음부피부총혈류관주량저우대조조(P <0.01);고환평균혈류관주수평화미혈관자률운동빈솔화진폭균현저저우대조조(P <0.01);당뇨병조소서생정상피수손,성숙생정세포감소;고환간질미혈관증다。고환간질미혈관내피세포 PECAM-1표체수평저우대조조(P <0.01)。결론:STZ 유도적 BALB/c 당뇨병소서고환미순배존재교다손해。
Objective:To investigate changes of testicular microcirculation in STZ induced BALB/c diabetic mice.Method:20 BALB/c mice were randomly divided into diabetic group (n=10)and control group (n=10).Di-abetic mice were treated with 40 mg/kg STZ for 5 consecutive days while control group was injected with citric acid buffer intraperitoneally.After one week,blood perfusion of lower abdominal-genital skin was evaluated by Moor LDLS,while testicular blood perfusion and microvascular vasomotion were detected by Moor VMS LDF after expo-sing.Followed cardiac perfusion,the testis of two group mice were prepared.HE staining was used to observe mor-phological and pathological of testicular micro-vessels,immunohistochemistry staining was employed to determine the expression of PECAM-1.Results:Compared with control group,diabetic mice had decreased lower abdominal-genital skin total blood perfusion (P <0.01),decreased average blood perfusion,frequency and amplitude of vaso-motion of testicular micro-vessels (P <0.01,respectively),damaged seminiferous epithelium,decreased matured spermatid,increased interstitial testicular capillaries and discontinuous expression of PECAM-1 (P <0.01).Conclu-sion:Testicular microcirculation impairs in STZ induced BALB/c diabetic mice.
