目的:建立动物模型观察脊髓损伤后的病理生理改变,寻找可直接判断脊髓损伤程度的方法,优选出治疗脊髓损伤的最佳方案。方法暴露大鼠L2-3脊髓后,于脊髓内注射0.5%布比卡因,评价大鼠行为学改变及脊髓形态学改变。观察并记录大鼠后肢行为学改变及脊髓神经元和周围组织的病理改变,评估脊髓损伤程度。结果与生理盐水注射组(对照组)相比,布比卡因注射组(观察组)大鼠脊髓损伤,表现为术后大鼠BBB评分降低,与对照组比较差异有统计学意义,病理HE染色可见脊髓神经元出现空泡变性及液化坏死。结论应用腰麻针进行大鼠脊髓穿刺并注射布比卡因的大鼠动物模型,具有稳定可控的脊髓损伤效果。

Objective To observe the pathophysiological changes after spinal cord injury by establishing the animal model,and to find the method that can directly determine the extent of spinal cord injury and optimize the best treatment for the injury. Methods After the exposure of the L2-3 spinal cord of each rat,0. 5% bupivacaine was injec-ted,and the changes in behavior and morphology were evaluated. The behavior changes of each rat′s hind limb and the histopathological changes of spinal cord neurons and the surrounding were observed and recorded to provide a evalua-tion of the degree of SCI. Results The spinal cord injury in rats of bupivacaine group was more serious compared with that of control group,the BBB score of postoperative rats in bupivacaine group was lower than that of control group, and the pathological HE staining of spinal cord neurons in bupivacaine group appeared empty degeneration and liquefac-tion necrosis. Conclusion The model of rats that received spinal needle puncture and injected with bupivacaine has a more stable and controllable effects on spinal cord injury.
