目的:研究N2a细胞分泌的外泌体对阿尔茨海默病( AD)模型小鼠β淀粉样蛋白( Aβ)浓度及斑块聚集的抑制作用,以期为临床治疗AD提供一种新的途径。方法将多步离心提取的外泌体持续注入表达β淀粉样前体蛋白( APP)的小鼠脑内,一段时间后检测Aβ浓度、Aβ老年斑块的形成以及APP小鼠的突触活性。结果经过持续外泌体注射的APP小鼠,Aβ浓度显著降低(P<0.05),Aβ斑块的形成也显著降低(P<0.05),而突触灵活性显著提高(P<0.05)。结论 N2a细胞分泌的外泌体对APP小鼠Aβ的表达有显著抑制作用。

목적:연구N2a세포분비적외비체대아이자해묵병( AD)모형소서β정분양단백( Aβ)농도급반괴취집적억제작용,이기위림상치료AD제공일충신적도경。방법장다보리심제취적외비체지속주입표체β정분양전체단백( APP)적소서뇌내,일단시간후검측Aβ농도、Aβ노년반괴적형성이급APP소서적돌촉활성。결과경과지속외비체주사적APP소서,Aβ농도현저강저(P<0.05),Aβ반괴적형성야현저강저(P<0.05),이돌촉령활성현저제고(P<0.05)。결론 N2a세포분비적외비체대APP소서Aβ적표체유현저억제작용。
Objective To study the effect of exosomes secreted by N2a cells on the concentration of β-amy-loid and the accumulation of plaque in Alzheimer′s mice. Methods The multi-step centrifugal extracted exosomes was continuously injected into the brain of AD mice,and the concentration of Aβ,formation of Aβplaques and synaptic ac-tivity in APP mice were detected. Results After continuous injection of exosomes in AD mice,the concentration of Aβ and the formation of Aβ plaques were significantly lower (P<0. 05) than those of control group,while the flexi-bility of synapse in AD mice significantly increased (P<0. 05) than that of control group. Conclusion Exosomes se-creted by N2a cells could significantly inhibitethe expression of Aβ in AD mice.
