目的:探讨乙型肝炎病毒X区基因变异与原发性肝癌的关系。方法39例乙型肝炎病毒( HBV)感染患者的组织标本中,慢性肝病(CHB)14例、肝细胞癌(HCC)25例。采用聚合酶链反应(PCR)方法扩增组织中HBV X区基因,并对PCR产物进行DNA测序,分析常见变异位点的变异情况。结果与CHB组相比,HCC组在X区发生插入或缺失变异及联合变异的位点及例数增多,并且A1762T与G1764A常发生双突变。 HBV X 区变异频率较高位点依次为:C1655T/G、A1605C/G、A1762T、G1764A、A1772B、A1645C、C1687A、G1776T。结论 HBV X区存在点突变、插入或缺失变异及联合变异,可能在HCC的发生发展过程中起重要作用。

목적:탐토을형간염병독X구기인변이여원발성간암적관계。방법39례을형간염병독( HBV)감염환자적조직표본중,만성간병(CHB)14례、간세포암(HCC)25례。채용취합매련반응(PCR)방법확증조직중HBV X구기인,병대PCR산물진행DNA측서,분석상견변이위점적변이정황。결과여CHB조상비,HCC조재X구발생삽입혹결실변이급연합변이적위점급례수증다,병차A1762T여G1764A상발생쌍돌변。 HBV X 구변이빈솔교고위점의차위:C1655T/G、A1605C/G、A1762T、G1764A、A1772B、A1645C、C1687A、G1776T。결론 HBV X구존재점돌변、삽입혹결실변이급연합변이,가능재HCC적발생발전과정중기중요작용。
Objective To investigate the relationship between multi?mutation of HBV X gene and hepatocellular carcinoma (HCC). Methods The live or liver cancer tissuses of 39 patients with HBV infected were divided into HCC group(25 cases) and chronic hepatitis B ( CHB ) group ( 14 cases ) . The HBV X gene were amplificated from above tissues by polymerase chain reaction ( PCR) , and the amplified products were sequenced directly. Furthermore, the mutation of common mutation site of HBV X gene were determined using soft of Bio Edit7?0 and ClustalW by computer. Results In HCC group, there were higher gene mutation frequency of insert/deletion mutation and combined mutation than those in CHB group. There was higher 1762T/G1764A double mutation frequency in HCC group than that in CHB group. The high frequency of mutation in HBV X gene were as follows:C1655T/G, A1605C/G, A1762T, G1764A, A1772B, A1645C, C1687A and G1776T. Conclusion The mutation of the HBV X region including point muta?tion, insertion/deletion mutation and combined mutation, which may play an important role in the occurrence and development of HCC.
