目的:建立裸鼠低位直肠癌淋巴结转移模型,检测E-钙黏蛋白在裸鼠低位直肠癌淋巴结转移模型中的表达,探讨其在直肠癌淋巴结转移过程中的作用。方法取对数生长期的人大肠癌细胞株SW480接种于裸鼠直肠黏膜下,建立裸鼠低位直肠癌淋巴结转移模型。按随机数字表法分为三组院直肠癌模型组(A组,40只,二甲基肼注射+SW480种植)、二甲基肼对照组(B组,40只,二甲基肼注射)和空白对照组(C组,40只,不作任何处理)。处死裸鼠,光镜下观察种植瘤组织和淋巴结转移情况,并采用免疫组化法检测种植瘤组织和转移淋巴结组织中E-钙黏蛋白表达水平。结果 A组裸鼠<1周时成瘤率为100豫,之后瘤结节逐渐长大。B组和C组裸鼠均未成瘤。A组裸鼠的肿瘤组织中E-钙黏蛋白阳性表达率(40.0%)明显低于肿瘤周边组织(82.5%),差异有统计学意义(字2=7.24,P<0.05)。 B组裸鼠的直肠黏膜组织中E-钙黏蛋白阳性表达率为80.0%,C组裸鼠的直肠黏膜组织中E-钙黏蛋白阳性表达率为85.0%,A组肿瘤组织以及B组、C组三组比较差异有统计学意义(字2=5.18,P<0.05),A组的E-钙黏蛋白阳性表达率明显低于B组和C组(字2=8.12、9.04,P<0.05)。发生侧方淋巴结转移的A组裸鼠种植瘤组织中E-钙黏蛋白阳性表达率为33.3%,明显低于未发生侧方淋巴结转移的裸鼠种植瘤组织(100.0%),差异有统计学意义(字2=10.28,P<0.05)。结论二甲基肼预处理后进行人大肠癌细胞株SW480接种制模法成功构建了裸鼠低位直肠癌淋巴结转移模型。 E-钙黏蛋白表达减弱可能在直肠癌发生、侵袭及淋巴结转移演进过程中起着重要作用。

목적:건립라서저위직장암림파결전이모형,검측E-개점단백재라서저위직장암림파결전이모형중적표체,탐토기재직장암림파결전이과정중적작용。방법취대수생장기적인대장암세포주SW480접충우라서직장점막하,건립라서저위직장암림파결전이모형。안수궤수자표법분위삼조원직장암모형조(A조,40지,이갑기정주사+SW480충식)、이갑기정대조조(B조,40지,이갑기정주사)화공백대조조(C조,40지,불작임하처리)。처사라서,광경하관찰충식류조직화림파결전이정황,병채용면역조화법검측충식류조직화전이림파결조직중E-개점단백표체수평。결과 A조라서<1주시성류솔위100예,지후류결절축점장대。B조화C조라서균미성류。A조라서적종류조직중E-개점단백양성표체솔(40.0%)명현저우종류주변조직(82.5%),차이유통계학의의(자2=7.24,P<0.05)。 B조라서적직장점막조직중E-개점단백양성표체솔위80.0%,C조라서적직장점막조직중E-개점단백양성표체솔위85.0%,A조종류조직이급B조、C조삼조비교차이유통계학의의(자2=5.18,P<0.05),A조적E-개점단백양성표체솔명현저우B조화C조(자2=8.12、9.04,P<0.05)。발생측방림파결전이적A조라서충식류조직중E-개점단백양성표체솔위33.3%,명현저우미발생측방림파결전이적라서충식류조직(100.0%),차이유통계학의의(자2=10.28,P<0.05)。결론이갑기정예처리후진행인대장암세포주SW480접충제모법성공구건료라서저위직장암림파결전이모형。 E-개점단백표체감약가능재직장암발생、침습급림파결전이연진과정중기착중요작용。
Objective To establish a nude mice low rectal cancer lymph node metastasis model and to detect the ex-pression of E-calcium in low rectal cancer lymph node metastasis in nude mice model, to discusse its role in the pro-cess of rectal cancer lymph node metastasis. Methods To establish a nude mice model of low rectal cancer lymph node metastasis by using people's congress colon cancer cell line SW480. There were three groups according to random num-ber table method: colorectal cancer model groups (group A, 40 cases, dimethyl hydrazine injection+ SW480 planting), dimethyl hydrazine in the control group (group B, 40 cases, dimethyl hydrazine injection) and blank control group (group C, 40 cases, do not make any processing). After Executing nude mice, the growth of tumor tissue and lymph node metastasis were observed, and the expression levels of E-calcium protein in tumor tissues and metastasis lymph node tissues were detected using immunohistochemical method. Results The rate of nude mice tumor of group A was 100% about 1 week, after tumor nodules gradually grew up. Group B and group C had no tumor. The positive expres-sion rate of E-calcium protein in tumor tissues of group A was 40.0%, was significantly lower than that in the tumor surrounding tissues of group A (82.5%), the difference was statistically significant (χ2=7.24, P< 0.05). The positive ex-pression rate of E-calcium protein in rectal mucosa tissues of group B was 80.0% and the positive expression rate of E-calcium protein in rectal mucosa tissues of group C was 85.0%. Comparison between tumor tissues of group A and group B, group C, the difference was statistically significant (χ2=5.18, P<0.05), the positive expression rate of E-calcium in group A was lower than that in group B and group C, the differences were statistically significant (χ2=8.12, 9.04, P<0.05). The positive expression rate of E-calci-um protein in tumor tissue of nude mice with lateral lymph node metastasis was 33.3%, was significantly lower than that in tumor tissue of nude mice without lateral lymph node metastasis (100.0%), the difference was statistically signifi-cant (χ2=10.28, P<0.05). Conclusion The nude mice model for low rectal cancer lymph node metastasis is successful-ly build by dimethyl hydrazine pedestrians colorectal cancer cell line SW480 vaccination moulding method. The lower expression of E-calcium protein may play an important role in the process of occurrence, invasion and lymph node metastasis in colorectal cancer.
