目的:对大鼠白蛋白姜黄素纳米粒子致SD大鼠的毒性和免疫原性进行考察。方法大鼠白蛋白姜黄素纳米粒子和姜黄素溶液,连续7 d通过尾静脉注射SD大鼠,同时设置对照(每组8只)。给药后第9天,将每组中4只大鼠处理,取心、肝、脾、肺和肾,用福尔马林浸泡。组织经包埋,切片, H&E染色,显微镜下,观察病理变化。21 d后,所有大鼠采血,酶联免疫吸附法检测给药组大鼠血清中是否产生针对大鼠白蛋白的抗体。结果与正常对照组相比,给药组大鼠的各脏器没有明显的病理变化。大鼠血清样本检测,没有检测出针对大鼠白蛋白的抗体。结论在试验周期内,白蛋白姜黄素纳米粒子不会导致受试动物的脏器发生显著的病理变化,且不会导致机体产生特异性抗体。

목적:대대서백단백강황소납미입자치SD대서적독성화면역원성진행고찰。방법대서백단백강황소납미입자화강황소용액,련속7 d통과미정맥주사SD대서,동시설치대조(매조8지)。급약후제9천,장매조중4지대서처리,취심、간、비、폐화신,용복이마림침포。조직경포매,절편, H&E염색,현미경하,관찰병리변화。21 d후,소유대서채혈,매련면역흡부법검측급약조대서혈청중시부산생침대대서백단백적항체。결과여정상대조조상비,급약조대서적각장기몰유명현적병리변화。대서혈청양본검측,몰유검측출침대대서백단백적항체。결론재시험주기내,백단백강황소납미입자불회도치수시동물적장기발생현저적병리변화,차불회도치궤체산생특이성항체。
Objective The rat albumin-curcumin nanoparticles induced toxicity and immunogenicity of SD rats was studied. Method Rat albumin -curcumin nanoparticles and curcumin solution were administered for 7 consecutive days via tail vein injection, and control groups with no drug administered were set (n=8).After the blood collection at 9 day, heart, liver, spleen, lung, kidneys of 4 rats in each group were harvested and treated with Faure Marin.After staining, the pathological changes were observed.After 21days, all the serum of rats were detected by enzyme -linked immunosorbent assay (ELISA).Results Compared with the normal control group, there were no obvious pathological changes in the organs of administered rats.No antibodies against rat albumin were detected.Conclusion In this study, the albumin nanoparticles can not cause the pathological changes of the organs of the animals studied, and no specific antibody is caused.
