目的:优化葛根的提取工艺参数,提高葛根素的提取率。方法在单因素初筛的基础上,以HPLC测定的葛根素含量为响应值,进行三因素三水平Box-Behnken试验设计,并利用Design Expert软件对试验数据进行分析,得到响应面数据并分析结果。结果乙醇浓度与提取时间、提取时间与料液比、乙醇浓度与料液比之间均存在着交互作用。葛根素的最优提取条件为乙醇浓度78%,提取时间为143 min,料液比为1∶10。结论优化的工艺提取条件简便,重现性良好,适用于葛根素的提取。

목적:우화갈근적제취공예삼수,제고갈근소적제취솔。방법재단인소초사적기출상,이HPLC측정적갈근소함량위향응치,진행삼인소삼수평Box-Behnken시험설계,병이용Design Expert연건대시험수거진행분석,득도향응면수거병분석결과。결과을순농도여제취시간、제취시간여료액비、을순농도여료액비지간균존재착교호작용。갈근소적최우제취조건위을순농도78%,제취시간위143 min,료액비위1∶10。결론우화적공예제취조건간편,중현성량호,괄용우갈근소적제취。
Objective To optimize the extraction technological parameters of Radix Puerariae in order to improve the ex-traction ratio of puerarin. Methods Based on the single factor preliminary screening ,with the puerarin content detected by HPLC as the response value,the three-factor and three-level Box-Behnken test design was carried on. The experimental data were ana-lyzed by using the Design-Expert software. Results The interaction action existed between the ethanol concentration and extrac-tion time,between the extraction time and solid-liquid ratio and between the ethanol concentration and solid-liquid ratio. The opti-mal extraction conditions of puerarin were the ethanol concentration of 78%,the extraction time of 143 min and the solid to liquid ratio of 1∶10. Conclusion The optimized technological extraction conditions are simple with good reproducibility and suitable for the extraction of puerarin.
