目的:探讨发育期大鼠癫痫持续状态对其海马内低氧诱导因子-1α(HIF-1α)和血红素加氧酶-1(HO-1)表达的影响。方法21 d SD大鼠随机分为2组:生理盐水组(NS组)、戊四氮(PTZ)致癫痫持续状态模型组(模型组)。1% PTZ(80 mg/kg)予大鼠腹腔注射诱导建立癫痫持续状态模型,生理盐水组予等量生理盐水腹腔注射,采用免疫组化和Western blot分别检测海马内HIF-1α、HO-1蛋白和阳性细胞数的表达。结果 HIF-1α、HO-1蛋白在正常生理状态下(NS组)均表达,而且表达平稳;在模型组存在明显的表达时程和表达高峰变化,HIF-1α蛋白1 h即有表达,4 h增多,8 h达高峰,然后逐渐下降;HO-1蛋白1 h少量表达,4~8 h逐渐增多,12 h达高峰,后渐下降。结论癫痫持续状态后,HIF-1α和HO-1表达升高可能是癫痫后的一个重要的脑保护机制。
目的:探討髮育期大鼠癲癇持續狀態對其海馬內低氧誘導因子-1α(HIF-1α)和血紅素加氧酶-1(HO-1)錶達的影響。方法21 d SD大鼠隨機分為2組:生理鹽水組(NS組)、戊四氮(PTZ)緻癲癇持續狀態模型組(模型組)。1% PTZ(80 mg/kg)予大鼠腹腔註射誘導建立癲癇持續狀態模型,生理鹽水組予等量生理鹽水腹腔註射,採用免疫組化和Western blot分彆檢測海馬內HIF-1α、HO-1蛋白和暘性細胞數的錶達。結果 HIF-1α、HO-1蛋白在正常生理狀態下(NS組)均錶達,而且錶達平穩;在模型組存在明顯的錶達時程和錶達高峰變化,HIF-1α蛋白1 h即有錶達,4 h增多,8 h達高峰,然後逐漸下降;HO-1蛋白1 h少量錶達,4~8 h逐漸增多,12 h達高峰,後漸下降。結論癲癇持續狀態後,HIF-1α和HO-1錶達升高可能是癲癇後的一箇重要的腦保護機製。
목적:탐토발육기대서전간지속상태대기해마내저양유도인자-1α(HIF-1α)화혈홍소가양매-1(HO-1)표체적영향。방법21 d SD대서수궤분위2조:생리염수조(NS조)、무사담(PTZ)치전간지속상태모형조(모형조)。1% PTZ(80 mg/kg)여대서복강주사유도건립전간지속상태모형,생리염수조여등량생리염수복강주사,채용면역조화화Western blot분별검측해마내HIF-1α、HO-1단백화양성세포수적표체。결과 HIF-1α、HO-1단백재정상생리상태하(NS조)균표체,이차표체평은;재모형조존재명현적표체시정화표체고봉변화,HIF-1α단백1 h즉유표체,4 h증다,8 h체고봉,연후축점하강;HO-1단백1 h소량표체,4~8 h축점증다,12 h체고봉,후점하강。결론전간지속상태후,HIF-1α화HO-1표체승고가능시전간후적일개중요적뇌보호궤제。
ObjectiveTo investigate the effects of the expression of HIF-1α and HO-1 in hippocampus of epileptic rats.MethodsSD rats aged 21 days were divided into two groups: normal saline group (NS), pentylenetetrazole (PTZ) induced status epilepticus (SE) group (model group). The status epilepticus rat model was established by intraperitoneal injection of 1% PTZ (80 mg/kg), and the same amount of saline were given intraperitonealy to rats of NS group. The hippocampus of rats were removed after SE and used to examine the expression of Notch-1 and HIF-1α using Western blot and immunohistochemistry methods.ResultsNotch-1 and HO-1 proteins were expressed lower stably in normal rats, and increased significantly in rats of model groups which began to increase at 1 h after SE and then peaked at 8 h.ConclusionAfter status epilepticus, expression increases of HIF-1α and HO-1 could be an important cerebral potential mechanism after seizure.
