目的: Notc h 信号在肠上皮细胞种系分化和引发肠腺瘤和肠癌中起着关键的作用。本文试图探讨Notch信号的配体JAG2在大肠癌中的作用。方法Real-Time PCR和Western blot分别检测JAG2特异性siRNA对JAG2 mRNA及蛋白表达的抑制率,细胞增殖实验和细胞侵袭实验观察siRNA对细胞增殖及侵袭能力的影响,人类肿瘤转移RT2 Profiler? PCR Array用于筛选JAG2下游靶基因,并应用特异性抑制化合物抑制靶蛋白活性。结果特异性siRNA对JAG2 mRNA及蛋白的表达具有抑制作用(P<0.05),对细胞增殖无明显影响(P>0.05),对细胞侵袭能力明显抑制(P<0.05),而组织蛋白K(CTSK)活性降低可能参与了这一过程。结论 JAG2基因可能对大肠癌细胞的侵袭转移能力起促进作用。

ObjectiveNotch signaling plays a critical role for specifying the intestinal epithelial cel lineage and for initiating colorectal adenomas and colorectal cancers (CRC). JAG2, a ligand of Notch signaling pathway, in colorectal cancer remains stil unclear.MethodsReal-Time PCR and Western blot were performed to determine the effects of JAG2 siRNA on mRNA and protein. Then, MTS cel proliferation assay and matrigel invasion assay were employed to test the functions of JAG2 siRNA on cel proliferation and invasion. Also, the human tumor metastasis RT2 Profiler? PCR Array were used for identify down-regulated gene by JAG2 siRNA in HCT-116 cels. Finally, pharmacological inhibition of CTSK activity was also perform to testify the function of CTSK on CRC cels.ResultsJAG2 siRNA markedly suppressed JAG2 mRNA and protein expression in HCT-116 and DLD-1 cels (P<0.05). CRC cel invasion but not proliferation were significantly attenuated by JAG2 siRNA, which may involve CTSK inactivation.ConclusionJAG2 gene may play a role in promotion of cel invasion in HCT-116 and DLD-1 cels.
