目的:筛选乳腺癌组织与癌旁正常乳腺组织之间的差异蛋白,寻找和鉴定与乳腺癌相关的蛋白质。方法选取新鲜乳腺浸润性导管癌组织及癌旁正常乳腺组织行双向凝胶电泳,得到的差异蛋白点进行图像分析,运用质谱分析对所筛选的差异蛋白点做质谱鉴定,根据检索软件Mascot计算得到蛋白的分值,输入NCBI数据库来获得蛋白质的信息。结果浸润性导管癌及癌旁正常乳腺组织分别获得(1040±15)个蛋白位点和(1003±17)个蛋白位点,差异在2倍以上共有28个蛋白位点,其中22个蛋白点表达上调,6个蛋白点表达下调,有23个蛋白点得到鉴定(蛋白得分大于70分),其中得分最高的为热休克蛋白27、70、90。结论热休克蛋白27、70、90在乳腺浸润性导管癌组织中呈现高表达状态,提示它们与乳腺癌密切相关,有可能成为乳腺浸润性导管癌的联合特异性标记物。

ObjectiveTo screen differential proteins between breast cancer tissues and adjacent normal breast tissues, and to search and identify proteins associated with breast cancer.Methods Proteins were extracted from fresh breast invasive ductal carcinoma and adjacent normal breast tissues. The two-dimensional gel electrophoresis was performed then protein spots were obtained and analyzed by image analysis. The differential protein spots were identified using mass spectrometry and the scores of proteins were calculated with retrieval software Mascot then were inputted into NCBI protein database in order to obtain information.Results(1 040±15) protein spots and (1 003±17) protein spots were obtained from invasive ductal carcinoma and adjacent normal breast tissues respectively. There were 28 protein spots with significant differences up to 2 fold between cancer and normal tissues, of which 22 protein spots up-regulated, 6 down-regulated and 23 protein spots identified whose protein scores were more than 70 points. Notably the highest ones were identified as heat shock protein 27, 70, 90.Conclusion Heat shock proteins 27, 70, 90 exhibit strong expression in invasive ductal breast carcinoma, suggesting that they are closely related to breast cancer, and may become a joint tumor-specific marker of breast invasive ductal carcinoma.
