Zhuxian Dan%Po1ycystic ovary syndrome%Hyperandrogenism
目的:观察助仙丹对多囊卵巢大鼠模型卵巢形态和性激素的影响。方法将60只大鼠随机分为2组,空白对照8只,另52只大鼠采用避孕硅胶棒联合皮下往射人绒毛膜促性腺激素方法进行造模,连续监测两个动情周期,选出造模成功大鼠30只,随机分为三组:助仙丹组(10只﹚、炔雌醇环丙孕酮片组(10只﹚、模型组(10只﹚,称重,记为用药前大鼠体质量,助仙丹组、炔雌醇环丙孕酮片组分别给予助仙丹配方颗粒和炔雌醇环丙孕酮片灌胃,模型组及空白对照组给予等量自来水灌胃,14天后再次称重,记为用药后体质量,后腹主动脉取血,检测血清雄激素浓度,并摘除双侧卵巢。利用单因素方差分析的统计学方法,观察各组大鼠用药前后体质量变化,卵巢质量变化,及血清雄激素浓度变化,并观察闭锁卵泡数量、颗粒细胞层数目,卵泡发育状态等卵巢形态变化。结果与模型组比较,炔雌醇环丙孕酮片组大鼠治闻前后体质量增加值明显降低,有显著差异(P<0.05﹚,炔雌醇环丙孕酮片组大鼠卵巢质量与模型组比较,有显著差异( P<0.05﹚;各组大鼠雄激素水平未见明显差异。助仙丹颗粒配方组大鼠卵巢切片与模型组大鼠相比:闭锁卵泡数量减少,卵巢颗粒细胞层数明显增加,可见正常发育阶段卵泡及成熟黄体组织。结论助仙丹可改善多囊卵巢综合征大鼠卵巢形态、促进正常排卵。

목적:관찰조선단대다낭란소대서모형란소형태화성격소적영향。방법장60지대서수궤분위2조,공백대조8지,령52지대서채용피잉규효봉연합피하왕사인융모막촉성선격소방법진행조모,련속감측량개동정주기,선출조모성공대서30지,수궤분위삼조:조선단조(10지﹚、결자순배병잉동편조(10지﹚、모형조(10지﹚,칭중,기위용약전대서체질량,조선단조、결자순배병잉동편조분별급여조선단배방과립화결자순배병잉동편관위,모형조급공백대조조급여등량자래수관위,14천후재차칭중,기위용약후체질량,후복주동맥취혈,검측혈청웅격소농도,병적제쌍측란소。이용단인소방차분석적통계학방법,관찰각조대서용약전후체질량변화,란소질량변화,급혈청웅격소농도변화,병관찰폐쇄란포수량、과립세포층수목,란포발육상태등란소형태변화。결과여모형조비교,결자순배병잉동편조대서치문전후체질량증가치명현강저,유현저차이(P<0.05﹚,결자순배병잉동편조대서란소질량여모형조비교,유현저차이( P<0.05﹚;각조대서웅격소수평미견명현차이。조선단과립배방조대서란소절편여모형조대서상비:폐쇄란포수량감소,란소과립세포층수명현증가,가견정상발육계단란포급성숙황체조직。결론조선단가개선다낭란소종합정대서란소형태、촉진정상배란。
Objective To investigate the effect of traditiona1 Chinese medicine modified Zhuxian Dan( TCM-ZXD﹚ on ovary tissues patho1ogy and improving sex hormone in rat mode1s of po1ycystic ovary syndrome( PCOS﹚. Methods 60 rats were random1y divided into 2 groups, 52 rats in patho1ogica1 mode1 group and 8 in norma1 contro1 group. Patho1ogica1 mode1 group was subcutaneous imp1anted of progesterone ge1 rod combined with HCG for bui1ding PCOS mode1s. According to the vagina1 smear, 30 successfu1 mode1 rats were se1ected and then random1y divided into three subgroups:TCM-ZXD group ( 10 rats﹚, Diane-35 group (10 rats﹚, mode1 group (10 rats﹚. A11 the rats were weighed, recorded as body weight before treatment. TCM-ZXD group and Diane-35 group were given the right amount of formu1a granu1es of TCM-ZXD and Diane-35, whi1e the mode1 group and the contro1 group were given the same amount of water. After 14 days of therapy, the rats were weighed again, recorded as body weight after treatment.
<br> B1ood was co11ected from abdomina1 aorta, then T in serum was determined. The weight of the ovary was recorded and ovarian morpho1ogy was observed. The resu1ts were ana1yzed by one-way anova. Result Compared with the mode1 group, the added va1ue of body weight of Diane-35 group before and after the treatment decreased significant1y ( P<0. 05﹚, there was a significant change in the Diane-35 group of the rats ovarian weight (P<0. 05﹚. There was no obvious difference in androgen 1eve1s in rats between these groups. According to the Ovary paraffin, TCM-ZXD group cou1d increase the 1ayers of ovarian granu1osa ce11s and the number of 1utea1 tissue, reduce the number of 1ocked fo11ic1es. Conclusion Zhuxian Dan can improve the ovarian morpho1ogy and promote ovu1ation in rats with PCOS.
