目的:探讨Slit2/Robo1信号通路在脑缺血后血管新生中的作用。方法:雄性SD大鼠48只,随机分为假手术组和模型1、3、7天组,每组12只。模型1、3、7天组采用线栓法制备大鼠永久性大脑中动脉缺血模型并分别于造模后第1、3、7天进行指标检测。应用Western blot方法检测缺血脑组织中Slit2蛋白的表达。应用RT-PCR方法检测缺血脑组织中Robo1、血管内皮生长因子(VEGF)mRNA的表达。利用CD105标记新生血管内皮细胞,计数大鼠脑组织中微血管密度( MVD)。结果:4组大鼠缺血脑组织中Slit2蛋白、Robo1和VEGF mRNA的表达及MVD差异有统计学意义(F=394.528、1058.653、898.938和142.753,P<0.001)。模型1、3、7天组Slit2蛋白、VEGF mR-NA表达水平及MVD均明显高于假手术组(P<0.05),并且逐渐升高(P<0.05);Robo1 mRNA的表达水平在3组间逐渐升高(P<0.05),但均低于假手术组(P<0.05)。模型组大鼠Slit2蛋白表达水平与VEGF mRNA表达水平、MVD呈正相关(r=0.974、0.926,P<0.001)。结论:Slit/Robo信号通路可能参与了大鼠脑缺血后的血管新生过程。

목적:탐토Slit2/Robo1신호통로재뇌결혈후혈관신생중적작용。방법:웅성SD대서48지,수궤분위가수술조화모형1、3、7천조,매조12지。모형1、3、7천조채용선전법제비대서영구성대뇌중동맥결혈모형병분별우조모후제1、3、7천진행지표검측。응용Western blot방법검측결혈뇌조직중Slit2단백적표체。응용RT-PCR방법검측결혈뇌조직중Robo1、혈관내피생장인자(VEGF)mRNA적표체。이용CD105표기신생혈관내피세포,계수대서뇌조직중미혈관밀도( MVD)。결과:4조대서결혈뇌조직중Slit2단백、Robo1화VEGF mRNA적표체급MVD차이유통계학의의(F=394.528、1058.653、898.938화142.753,P<0.001)。모형1、3、7천조Slit2단백、VEGF mR-NA표체수평급MVD균명현고우가수술조(P<0.05),병차축점승고(P<0.05);Robo1 mRNA적표체수평재3조간축점승고(P<0.05),단균저우가수술조(P<0.05)。모형조대서Slit2단백표체수평여VEGF mRNA표체수평、MVD정정상관(r=0.974、0.926,P<0.001)。결론:Slit/Robo신호통로가능삼여료대서뇌결혈후적혈관신생과정。
Aim:To explore the relationship between Slit 2/Robo1 signaling pathway and angiogenesis after ischemia . Methods:Forty-eight male SD rats were randomly allocated into sham operation group and D 1,D3,D7 model groups(n=12).The D1,D3,D7 model groups were to establish cerebral ischemia model by the suture method .Western blot was used to analyze the Slit2 expression in infarction brain tissue and RT-PCR was used to detect the expressions of Robo 1 and VEGF mRNA.Microvessel density ( MVD) was calculated through marking the vessel endothelial cells with CD 105.Results:There were significant differences in the Slit 2 protein, Robo1 and VEGF mRNA expressions ,and MVD among the 4 groups (F=394.528,1 058.653,898.938 and 142.753,P<0.001).The expressions of Slit2 protein, VEGF mRNA and MVD in D1,D3,D7 model groups were higher than that of the sham group , and increased by turn(P<0.05); the Robo1 mRNA expression were increased by turn(P<0.05),but still lower than that of the sham operation operation group .In the 3 model groups, the Slit2 protein expression was positively correlated with VEGF mRNA and MVD (r=0.974,0.926,P<0.001). Conclusion:Slit2/Robo1 signaling pathway may be involved in the process of angiogenesis after focal cerebral ischemia .
