目的 探讨大肠癌中P38-MAPK对CDX2表达的影响和CDX2的表达变化与患者预后之间的关系.方法 收集大肠癌患者病理组织共74例,采用qRT-PCR、Western印迹、免疫组化等技术检测P38-MAPK、CDX2在肿瘤组织、癌旁正常组织中的表达.收集患者的临床资料并进行术后随访,研究患者组织样本中P38-MAPK、CDX2表达的关系及CDX2的表达变化与患者术后生存之间的关系.结果 大肠癌患者肿瘤组织中P38-MAPK、CDX2均为低表达,两者表达变化呈正相关(r=0.4656,P<0.05).CDX2的表达变化与患者预后显著相关且(r=0.5148,P<0.05) CDX2高表达组患者具有更长的术后生存期.结论 大肠癌中P38-MAPK低表达导致了CDX2的表达降低.CDX2的表达变化可以用来评估患者的术后生存并可能成为大肠癌治疗的新靶点.

Objective To investigate the relationship between P38-MAPK and CDX2 expression and to explore the correlation between expression of CDX2 and the survival of colorectal cancer patients.Methods From January 2008 to January 2010,a total of 74 colorectal cancer patients were included and tumor tissues and their adjacent normal tissues were collected.qRT-PCR,Western blot,immunohistochemical staining were applied to detect the expression of P38-MAPK and CDX2.Furthermore,the postoperative survival of the tumor patients was also studied.Results The expression of P38-MAPK and CDX2 of tumor tissues was lower than that of the adjacent normal tissues.P38-MAPK expression was positively associated with CDX2 expression (r =0.4656,P < 0.05).CDX2 correlated with the postoperative survival of the tumor patients (r =0.5148,P < 0.05).The postoperative survival of high CDX2 patients was longer than that of low CDX2 patients.Conclusion The decreased expression of P38-MAPK results in decreased expression of CDX2 and thus,the CDX2 expression could be used to predict the postoperative survival of colorectal cancer patients and might be a novel antitumor therapeutic target.
