目的:观察骨疏颗粒对去卵巢骨质疏松大鼠骨密度及血清细胞因子的影响,探讨骨疏颗粒防治骨质疏松症的可能作用机制。方法将3月龄SD雌性大鼠40只,按随机数字表法分为空白组、模型组、戊酸雌二醇组及骨疏颗粒组。空白组只行假手术,其余3组用去大鼠双侧卵巢复制绝经后骨质疏松症的模型。术后3天分别给戊酸雌二醇及骨疏颗粒灌胃,12周后检测血清白细胞介素(interleukin,IL)1β、IL-6、肿瘤坏死因子(tumor necrosis factor,TNF)-α及左股骨骨密度。结果(1)与空白组比较,模型组骨密度显著下降,血清 IL-1β及IL-6显著增加(P<0.01),TNF-α增加(P<0.05);(2)与模型组比较,戊酸雌二醇组骨密度显著增加(P<0.01),血清 IL-1β显著降低(P<0.01),IL-6及TNF-α均降低(P<0.05);骨疏颗粒组骨密度增加(P<0.05),血清IL-1β、IL-6及TNF-α均降低(P<0.05)。结论骨疏颗粒对去卵巢骨质疏松大鼠有治疗作用,其作用机制可能是通过降低血清中IL-1β、IL-6及TNF-α细胞因子水平,从而抑制骨吸收。

목적:관찰골소과립대거란소골질소송대서골밀도급혈청세포인자적영향,탐토골소과립방치골질소송증적가능작용궤제。방법장3월령SD자성대서40지,안수궤수자표법분위공백조、모형조、무산자이순조급골소과립조。공백조지행가수술,기여3조용거대서쌍측란소복제절경후골질소송증적모형。술후3천분별급무산자이순급골소과립관위,12주후검측혈청백세포개소(interleukin,IL)1β、IL-6、종류배사인자(tumor necrosis factor,TNF)-α급좌고골골밀도。결과(1)여공백조비교,모형조골밀도현저하강,혈청 IL-1β급IL-6현저증가(P<0.01),TNF-α증가(P<0.05);(2)여모형조비교,무산자이순조골밀도현저증가(P<0.01),혈청 IL-1β현저강저(P<0.01),IL-6급TNF-α균강저(P<0.05);골소과립조골밀도증가(P<0.05),혈청IL-1β、IL-6급TNF-α균강저(P<0.05)。결론골소과립대거란소골질소송대서유치료작용,기작용궤제가능시통과강저혈청중IL-1β、IL-6급TNF-α세포인자수평,종이억제골흡수。
Objective To observe Gushu powder treating rats with ovriectomy osteoporosis, and explore the mechanism of Gushu powder protecting and treating osteoporosis. Methods According to random number table, forty SD female rats of three months were randomly divided into four groups: sham group, model group, estradiol valerate group, and Gushu powder group. The sham group only got sham-operation and the other three groups were ovriectomized respectively. Three days later after ovriectomy, the rats were given estradiol valerate ( 0. 5 mg/kg ) and Gushu powder ( 10 mL/kg ) by intragastric administration for twelve weeks. Bone minereal density(BMD) was detected with dual-energy X-ray ab-sorptiometry. Serum interleukin-1 beta(IL-1β),interleukin-6(IL-6) and tumor necrosis factor alpha(TNF-α) were detected. Results ( 1 ) Compared with the sham group, the BMD significantly decreased (P<0.01),IL-1β and IL-6 significantly increased(P<0. 01), and TNF-α increased in model group (P<0.05). (2)Compared with model group,BMD in Bujiale group dramatically increased(P<0.01), IL-1β significantly decreased(P<0. 01),IL-6 and TNF-α decreased(P<0. 05). While in Gushu powder group, BMD in Bujiale group increased(P<0. 05);IL-1β, IL-6 and TNF-α decreased(P<0.05). Conclusions Gushu powder may prevent and treat osteoporosis through reducing sereum IL-1β, IL-6 and TNF-α which can depress bone absorption.
